摘 要:目的研究羧甲基壳聚糖(CM-COS)对阿霉素所致红细胞膜损伤的影响。方法用阿霉素作用红细胞膜,建立膜损伤实验模型。以红细胞形态学、细胞溶血率、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量、细胞膜流动性、封闭能力等为指标,观察羧甲基壳聚糖对红细胞膜损伤的保护作用。结果阿霉素能引起红细胞溶血率和MDA含量增加,SOD活性降低、膜流动性以及膜封闭度下降。而经过一定量的羧甲基壳聚糖(400,600,800μg·mL-1)预处理后红细胞溶血率和MDA含量明显减少,SOD活性、膜流动性和膜封闭度提高。结论羧甲基壳聚糖对阿霉素引发的红细胞膜损伤具有一定的保护作用。Objective To investigate the protect effects of carboxymethyl chitosan on the damaged erythrocyte membrane induced by doxorubicin. Methods Doxorubicin was used to induce damage to erythrocyte membrane at first. Then, carboxymethyl chitosan was taken to intervene this damaging effect and the erythrocyte morphology, the rate of hematolysis, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity of erythrocyte, erythrocyte membrane fluidity and blocking degree were determined to evaluate the protective effect of carboxymethylchitosan on this damaged erythrocyte membrane. Results The rate of bematolysis and erythrocyte MDA content were increased significantly and SOD activity, membrane fluidity and blocking degree of the erythrocyte were decreased owing to the use of doxorubicin. When the carboxymethyl chitosan at doses of 400, 600, 800μg·mL^-1 were pre-treated, the rate of hematolysis and MDA content of erythrocyte were decelerated obviously and the erythrocyte SOD activity, erythrocyte membrane fluidity and blocking degree were accelerated obviously. Conclusion Carboxymethyl chitosan showed significant protective effects on erythrocyte membrane damaged by doxorubicin.
