摘  要:目的研究镍矿尘对细胞增殖及DNA损伤的影响,为镍矿尘致癌性的实验研究提供科学依据。方法小鼠肺成纤维细胞(NIH/3T3)为靶细胞,以某选矿车间镍矿尘为受试物,采用浓度为12.5、25.0、50.0、100.0、200.0μg/ml进行细胞染毒,采用四甲基偶氮噻唑蓝(MTT)法检测细胞生长抑制率;采用浓度25.0、50.0、100.0μg/ml染毒,同时设PBS阴性对照组和阳性对照组(紫外光照射180 s)采用单细胞凝胶电泳技术检测细胞DNA损伤程度。结果镍矿尘能够使细胞生长受到抑制,呈剂量-反应关系,染毒24 h,100.0μg/ml浓度组细胞生长抑制率为63.1%,IC50为57.9μg/ml。镍矿尘还能使细胞DNA发生断裂,染毒组的细胞拖尾率、彗星细胞的Olive尾距、ExtentTM均增高,与阴性对照组比较,差异均有统计学意义(P<0.05)。结论选矿车间镍矿尘能够抑制细胞生长,具有对DNA损伤的作用。Objective To study the effects of nickel dust on cell multiplication and DNA damage and to provide scientific criteria for carcinogenieity experiment of nickel dust. Methods The NIH/3T3 cells of mice were taken as the target cell, and nickel dust from dressing workshop as the experiment material. The ceils were exposed to 12.5, 25.0, 50.0, 100.0 and 200.0 μg/ml. The ceil inhibition ratio was detected by MTT colorimetric assay. The cells were exposed to 25.0, 50.0 and 100.0 μg/ml; at the same time the PBS negative control and positive control groups(radiate under ultraviolet light for 180 s) were set. The DNA damage was determined with single cell gel electrophoresis(SCGE)technique. Results The nickel dust could inhibit the cell growth showing dose-response relationship. When exposing for 24 h, the cell inhibition ratio of 100.0μg/ml group was 63.1% and IC50 was 57.9 μg/ml. Nickel dust could also break the cell DNA into fragments. In positive group all the percentage of tailed cells, olive tail moment and extent TM increased, the difference was significant in comparison with those of the negative group (P〈0.05). Conclusions The nickel dust from the ore dressing shop could inhibit the cell growth, and damage the DNA.
