益气养阴方对CAM移植瘤组织MMP-2表达的影响

摘　　要：目的研究益气养阴方对肿瘤诱导鸡胚尿囊膜(CAM)血管生成及对CAM移植瘤细胞细胞基质金属蛋白酶-2(MMP-2)表达的效应。方法将大鼠20只随机分为2组,即药物组和对照组,各10只。药物组予益气养阴方水煎液灌胃,对照组予相同容积的0.9%氯化钠注射液灌胃,制备含药血清和对照血清。制备接种肿瘤细胞的CAM模型,分别予对照血清和含药血清各100μL,观察肿瘤细胞诱导血管生长情况,于解剖显微镜下观察CAM血管,计数血管数目和血管的面积,观察CAM移植瘤组织生长情况,并取肿瘤组织进行组织学观察,应用免疫组织化学及其组织形态学定量分析方法对MMP-2在CAM移植瘤中的表达进行研究。结果含药血清组CAM血管数目、血管面积均低于对照血清组,2组比较差异有统计学意义(P<0.01);含药血清组MMP-2灰度值高于对照血清组,2组比较差异有统计学意义(P<0.01)。结论益气养阴方对肿瘤诱导的CAM血管生成有明显抑制作用,降低MMP-2的表达可能是其抑制肿瘤血管生成的主要机制之一。Objective To study the role of reinforcing qi and nourishing yin on angiogenesis and on matrix metalloproteinase 2 （MMP2） in tumor cells transplanted on chick embryo chorioalantoic membrane （CAM）. Methods 20 rats were randomly divided into two groups. Rat in treatment group （ n = 10） were filled with water solution of reinforcing qi and nourishing in the stomach. Rat in control group （ n = 10） were lavagen with isometrical 0.9% NaC1 solution. Drug - treated serum and control serum were prepared. Chicken CAM that tumor cells transplanted was adopted. 100p^L Drug - treated serum and control serum were added. The number and area of microvessels of CAM were observed under anatomical microscope. The expression of MMP2 in tumor cells transplanted on CAM was investigated by immunohistochemical and morphological analytics methods. Results The number and area of microvessels of CAM I treatment group were lower than those in control group （ P 〈 0.01 ）. The expression of MMP2 in treatment group was lower than that in the control group （ P 〈 0.01 ）. Conclusion Reinforcing qi and nourishing yin has inhibitory effects on angiogenesis induced by tumor cells in CAM. Its mechanism may be associated with the down - regulating of MMP2 expression.
