摘 要:目的观察苦参碱对豆蛋白A(Concanavalin A,Con A)所致免疫性肝损伤小鼠肝组织Fas、FasL表达的影响,探讨其抗肝损伤的作用机制。方法70只NIH小鼠随机分为5组,分别为正常对照组,模型组、苦参碱高剂量组、低剂量组和联苯双酯治疗组。除正常对照组外,其他组于实验首日静脉注射Con A20mg/kg后,苦参碱高剂量组、低剂量组均采用尾静脉注射给药,联苯双酯灌胃口服,1次/d,连续5d,末次给药后4h,再次尾静脉注射Con A20mg/kg,8h后检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,观察肝组织病理学变化和Fas、FasL的表达。结果苦参碱高、低剂量组小鼠血清ALT、AST活性均明显低于模型组,差异有显著性意义(P<0.01),苦参碱各剂量组肝脏病理改变明显减轻,肝组织Fas、FasL的表达明显减弱,与模型组比较,差异均有显著性意义(P<0.01)。结论苦参碱对ConA所致小鼠免疫性肝损伤具有较好的保护作用,抑制肝组织中Fas、FasL的表达,从而减轻Fas/FasL系统介导的肝细胞凋亡,是其抗肝损伤的作用机制之一。Objective To observe effects of matrine on the expression of Fas and FasL in Liver tissue in mice with immune hepatic injury induced by eoncanavalin A (Con A),and inspect its mechanism of anti-liver injury. Methods Seventy NIH mice were randomly divided into 5 groups: normal control group(group A),model group (group B),high dosage group of Matrine (group C),low dosage group of Matrine (group D) and bifendate group (group E).Excepts those in group A,mice in other groups were injected with Con A (dose of 20mg/kg) via the tail vein at the first day of experiment. Group C and group D were injected via the tail vein with Matrine at does dose of 25mg/kg and 12.Smg/kg,respectively. Group E were orally administered with bifendate at the does dose of 150mg/kg.All the drugs were given once daily for 5 days consecutively. Finally the mice were injected with Con A once again at the same dosage 4 hours after administered of drug. Blood samples for determineing activity of alanine aminotransferase (ALT) and asparate aminotransferase (AST) were collected at the 8th hour after Con A administration. Histopathological examination was performed for liver tissue,the experssion of Fas and FasL in hepatic tissue was detected by immunohistochemical method. Results Activity of ALT and AST in group C and group D were all obviously lower than that of group B (P〈0.01),hepatic histopathological changes were alleviated in group C and group D, the experssion of Fas and FasL in liver tissue of group C and group D were all obviously attenuated comparing with that of group B(P〈0.01 ). Conclusion Matrine has a good protective effects on hepatic immune injury in mice induced by Con A and the mechanism is related to suppression of experssion of Fas and FasL in liver tissue.
