摘 要:[目的]提高大肠杆菌DH5α转化效率。[方法]在其他条件一致的情况下,将5μl的质粒或连接产物加入含有100μl的感受态0.2 ml PCR管和1.5 ml的离心管中,0.2 ml PCR管设置热休克时间梯度和1.5 ml的离心管热休克时间90 s,对大肠杆菌DH5α转化效率进行了比较。[结果]0.2 ml PCR管转化效率高于1.5 ml的离心管,且0.2 ml PCR管热休克30 s时最高。[结论]采用此方法提高了大肠杆菌DH5α的转化效率。[ Objective] The research aimed to improve the transformation efficiency of E. coli DH5α. [ Method ] Under the same other conditions, 5 μl plasmid or legation product was added into 0.2ml PCR tube and 1.5 ml centrifuge tube with 100 μl competent in them. In different thermal shock time, transformation efficiency of E. coli DH5α was compared. [ Result] Transformation efficiency with 0.2 ml PRC reformer tube is better than that with 1.5 ml centrifuge tube and the 0.2 ml PCR reformer tube with 30 seconds' thermal shock time gives the best performance of transformation efficiency. [ Conclusion ] Transformation efficiency of E. coli DH5α is effectively improved by this method.
