摘 要:目的观察纤维连接蛋白(Fn)及转化生长因子βⅡ型受体(TGF-βRⅡ)在小鼠肾小体发育中的时空性表达。方法采用免疫组织化学技术结合体视学方法测定不同胚龄(E,12、14、16和18d)及生后日龄(P,1、3、7、14、21、28和42d)小鼠肾小体Fn和TGF-βRⅡ的表达及其含量变化。结果Fn在除逗号小体以外的各期肾小体基底膜处均有表达,TGF-βRⅡ在各期肾小体内均有表达,且两者的表达量均随着肾小体的发育逐渐增加。结论Fn和TGF-βRⅡ在小鼠肾小体发育中的表达具有一定的时空性,推测两者在小鼠肾小体发育中起一定作用。Objective To observe the chronological and spatial expression of fibronectin (Fn) and TGF-βRⅡ in developing mouse glomeruli. Methods The expressions of Fn and TGF βRⅡ were examined by immunohistochemieal and stereological method in embryos (including fetal age of 12, 14, 16 and 18d) and postnatal mouse kidneys (including the age of 1, 3, 7, 14, 21, 28 and 42d). Results Immunohistochemical results showed that at fetal age of 14d, Fn was stained in S-shaped corpuscles, but not stained in the comma-shaped corpuscles. After fetal age 16d, Fn was expressed in all stages of glomeruli except the corona-shaped corpuscles. TGF βRⅡ was stained in all stages of glomeruli from fetal age 14d. Fn and TGF-βRⅡ were localized respectively in basement membrane and cell plasma of glomeruli. Stereological analysis showed that the expression of Fn was increased gradually in all stages of glomeruli with renal development, except in the comma-shaped corpuscles; also, the expression of TGF βRⅡ increased gradually in all stages of glomeruli with renal development. Conclusion The expressions of Fn and TGF βRⅡ in embryonic and postnatal mouse kidney show chronological and spatial sequence, implying that they might play a role in the development and maturation of renal glomeruli of mouse.
