壮肝逐瘀煎对肝纤维化大鼠转化生长因子β_1表达的影响

摘　　要：目的:观察壮肝逐瘀煎对肝纤维化大鼠血清及肝组织中TGF-β1表达的影响,探讨本方治疗肝纤维化的作用机理。方法:将100只Wistar大鼠随机分为正常对照组20只、观察组80只,参照相关文献复制大鼠肝纤维化模型,造模过程中,于第4周末随机处死正常对照组及观察组大鼠各5只,证实肝纤维化形成后,将观察组其余大鼠随机分为病理模型组、壮肝逐瘀煎大、中、小剂量组及秋水仙碱对照组,分别予生理盐水、壮肝逐瘀煎大、中、小剂量及秋水仙碱灌胃,日1次;12周后处死大鼠,应用ELISA法检测血清TGF-β1含量的改变,同时采用免疫组化观察肝脏组织中TGF-β1的表达水平。结果:壮肝逐瘀煎能降低大鼠肝纤维化血清中TGF-β1的表达,拮抗肝纤维化大鼠肝组织中TGF-β1的分泌。结论:壮肝逐瘀煎能有效抗大鼠肝纤维化,其作用机制是通过抑制TGF-β1的活性来实现的。Objective：To obsreve the effects of Zhuanggan Zhuyu Decoction on expression of TGF-β1 in blood serum and liver tissues of rats with hepatic fibrosis,and to probe the mechanism of the prescription in treating bepatic fibrosis.Methods：100 Wistar rats were randomly divided into control group（20 cases ）and observaton group（80cases）.During the modeling of rats with hepatic fibrosis according to the relative references,5 rats in each group were randomly killed at the end of the 4th week to verify the formation of hepatic fibrosis.Other rats in observation group were randomly divided into model group,high dose group,medium dose group and low dose group of Zhuanggan Zhuyu Deconction and Colchicine control group,they were respectively given gastric per fusion of normal saline,high dose group,medium dose group and low dose of Zhuanggan Zhuyu Decoction and colchicine once a day,The rats were killed 12 weeks later to detect the content change of blood serum TGF-β1 with ELISA method,and to observe the expreasion level of TGF-β1 of liver tissues.Results：Zhuanggan XZhuyu Decoction can decrease the expression of TGF-β1 in blood serum and fight against the secretion of TGF-β1 in liver tissues of rats with hepatic fibrosis.Conclusion：Zhuanggan Zhuyu Decoction can effectively resist the hepatic fibrosis,which is realized by inhibiting the activity of TGF-β1.
