摘 要:研究脑内交感抑制系统中中缝隐核(NRO)至中脑导水管周围灰质腹侧部(vPAG)多突触通路的儿茶酚胺中间神经元定位。方法在戊巴比妥钠麻醉的大鼠,应用原位杂交组织化学法检测电刺激激活NRO对酪氨酸羟化酶(TH)mRNA表达的影响,同时采用辣根过氧化物酶(HRP)逆行标记与免疫组化双标法以观察投射至vPAG的脑干儿茶酚胺能神经元分布。结果激活NRO18~20h后引起脑干延髓头端腹外侧区(rVLM)、孤束核(NTS)、蓝斑(LC)、中缝背核的THmRNA的表达增加;HRP/TH双标细胞分布于rVLM(C1)、NTS、LC、黑质网带、室周灰质等区。结论NRO至vPAG途径的肾上腺素、去甲肾上腺素神经元主要位于rVLM的C1区、NTS、LC。おURPOSE To examine the distribution of catecholamine neurons in the pathway from nucleus raphe obscurus (NRO) to the ventral periaqueductal grey (vPAG). To study the expression of tyrosine hydroxylase (TH) mRNA actvated by stimulation of NRO and the distribution of catecholamine neurons projecting to vPAG.METHODS Experiments were performed in rats anaesthetized with sodium pentobarbital. Perfusion was performed 18~20 hours after stimulating NRO for 1 hour. The brainstem sections were then prepared for visua ̄lization of TH mRNA positive cells with in situ hybridization. Furthermore, after iontophoretic application of HRP into vPAG, the brainstem sections were processed for visualization of retrogradely labeled cells and immu ̄nodetection of TH with ABC-DAB immunohistochemical techniqne.RESULTS It was observed that stimulation of NRO resulted in an increase in expression of TH mRNA in C1 of rostral ventrolateral medulla(rVLM), nucleus tractus solitarius(NTS), locus ceruleus(LC) and dorsal raphe(DR). Double-labeled neurons were observed in C1 of rVLM, NTS, LC, substantia nigra reticularis (SNR) and periventricular grey.CONCLUSIONS The results reveal that the cell bodies of adrenalinergic and noradrenalinergic neurons projecting to vPAG from NRO are located mainly in C1 of rVLM, NTS and LC.
