摘  要:目的呼吸道合胞病毒(RSV)F蛋白是RSV感染免疫中最重要的病毒蛋白,为了解我国RSV地方株F蛋白的基因状况和变异特征,随机选取北京、广州、长春和河北四个地区具有不同流行特征的RSV地方株(A亚型)5株,进行RSVF蛋白全基因的核苷酸序列分析。方法以提取的病毒mRNA为模板进行RT-PCR扩增、目的基因的克隆及序列测定,对地方株及原型株的序列进行比较分析。结果地方株F蛋白基因与原型株A2株有很高的同源性,核苷酸全序列的同源性为95.1%~96.1%,氨基酸同源性为96.7%~97.4%。核苷酸有义突变率为22.6%~25.9%。3非编码区的核苷酸序列比蛋白编码区变异显著。河北地方株(E73株)在3非编码区有6个核苷酸的插入。F2亚单位的氨基酸变异高于F1亚单位。在北京地方株(ZHS13株)F1亚单位内,由具有中和能力单克隆抗体所识别的抗原表位区中存在一个氨基酸的变异。结论我国RSV地方株与原型株之间的F蛋白基因尽管存在一定的变异,但仍有很高的同源性。地方株间F蛋白的核苷酸、氨基酸变异的位置及形式很相似,提示我国RSV的不同流行特征可能并非由于F蛋白的基因变异所致。Objective The fusion (F) protein seems a major target for the immune system after respiratory syncytial virus (RSV) infection. The field strains of Beijing,Guangzhou,Changchun and Hebei which have different epidemiological characteristics of RSV infection are selected at rendom. Methods The complete nucleotide sequences of the F protein gene were determined from cDNA clones that had been amplified by RT PCR and cloned into PTZ 18R plasmid vector and compared with those of their previously published counterparts of A 2 strain. Results The data indicated that the nucleotide sequences of the five RSV strains F mRNA had 95.1%—96.1% overall nucleotide identity with the previously published strain A 2 F mRNA sequence.The deduced amino acid sequences of the five strains and A 2 F proteins had 96.7%—97.4% overall amino acid identity. The accumulation of nucleotide changes of the long 3 non translated end were more frequent than those of the F mRNAs that coded for protein.There are six nucleotides insertion in the noncoding 3 end of the E73 strain.The indicated preferential accumulation of amino acid changes in the F 2 over the F 1 subunit is noteworthy.And there is an amino acid substitution located in the antigenic site of the large F 1 subunit of the ZHS13 strain,recognized by neutralizing monoclonal antibodies (McAb) directed against the F protein of human RS virus. Conclusion Although there are some differencs between the A 2 and the field strains in the F gene,the nucleotide identity is still high.The site and pattern of the nucleotide and amino acid substitutions among these field strains are similar,which indicates that the variability of the F protein may not result in the difference of the epidemiological characteristics among the field strains.
