高效液相色谱法测定颈康胶囊中葛根有效成分葛根素的含量

摘　　要：目的建立高效液相色谱法(HPLC)测定中成药颈康胶囊中葛根的主要成分葛根素的含量测定方法。方法采用Betasil(4.6mm×250mm,5μm)色谱柱;检测波长:250nm;流动相:甲醇-水(25∶75);流速:1.0mL/min。结果葛根素在0.14～0.694μg范围内具有良好的线性关系,相关系数r=0.999853,平均回收率为102.51%,RSD=1.24%。结论该方法灵敏度高、专属性强、重现性好,并通过对数批样品含量进行考察,可作为制剂质量控制指标之一。Objective To establish the method of determination of puerarin content,the main ingredient of Chinese herbal patent, Jing Kang Jiaonang with high performance liquid chromatography （HPLC）. Methods Betasil chromatographic column（4.6 mm×250 mm,5 μm）was adopted,the detective wavelength was 250 nm,mobile phase was methanol - water（25： 75） , and the flow rate was 1 mL/min. Results Good linear relation of puerarin was obtained in the range of 0.14 μg to 0.694 μg. Correlation coefficient r = 0. 999 853,the average recovery rate was 102.51% ,and RSD = 1.24%. Conclusion The method is high in sensitivity,strong in specialty and good in reproducibility. By the observation of puerarin contents of several batches of samples,it figures out that this method can be taken as one of the indexes to evaluate the quality of the preparation.
