人血清生长激素结合蛋白活性的快速测定法及其初步临床应用

摘　　要：利用葡聚糖包裹活性炭分离技术，建立了人血清生长激素结合蛋白（GH-BP）活性快速测定法，并在方法学方面进行了探讨。本法批内CV为4.9％，批间CV为9．6％。结果显示正常男性、女性、儿童及妊娠后期的孕妇的GH-BP水平之间无显著性差异（P＞0．05）；而肝硬化患者和一些生长激素（GH）分泌正常的矮小患儿的GH-BP水平明显降低（P＜0001）。提示人血清GH-BP的测定，对GH不缺乏的矮小儿的临床诊断以及与生长激素受体（GHR）改变有关的疾病研究具有十分重要的意义。In this report we describe a simple and rapid assay for measuring GH-BP in human serum, based on the dextran- coated charcoal technique. The assay for measuring GH-BPhas been studied in time, temperature, serum concentration, concentration of separate dose, implications of endogenous hGH and quality control. The intra-and inter-assay coeficients of variation (CV) were 4. 9 % and 9. 6 %, respectively. The results indicate that the mean GH-BP activities in 50μL sera from normal adult men, adult women, children and pregnancy were 12. 89%±1. 60%, 12. 25%± 1. 91 %, 12. 34% ± 1. 54% and 12. 46% ±1. 26%, respectively. Therewas no significant difference. Levels of GH－BP were lower in short－statured children with normal GH secretion (8. 24 % ±2. 2 % ) and patients with liver cirrhosis (7. 55 % ±1.79 % ). The results indicate that the rapid assay for GH- BP activity in human serum is very useful for the studyof GH receptors and short-statured children diagnosis with normal GH secretion.
