摘 要:[目的]探讨紫锥菊多酚植物化学物质对因束缚-浸水应激引起的大鼠应激性胃溃疡的影响。[方法]将50只健康SD大鼠(雌雄各半)随机分为5组,即紫锥菊多酚提取物组、红景天苷组、西米替丁对照组、生理盐水模型组和生理盐水对照组;连续灌胃给药7 d,末次给药后禁食不禁水;采用"束缚-浸水"应激法制备大鼠应激性胃溃疡模型后选取胃、脾脏,测定其溃疡指数,脾指数,大鼠胃黏膜中一氧化氮合酶(NOS)活力。[结果]紫锥菊多酚提取物组的溃疡指数为(15±2)分与生理盐水模型组的溃疡指数(65±4)分比较数值明显降低,差异有显著性意义(P<0.01);该组脾指数为(5.425±0.548),与生理盐水模型组的脾指数(2.323±0.392)比较数值明显增高,差异有显著性意义(P<0.01);该组大鼠胃黏膜腺胃部和胃底部的NOS活力分别为(56.249±3.473)U/mgprot和(53.852±6.146)U/mgprot,与生理盐水模型组的(28.240±4.956)U/mgprot和(27.173±4.244)U/mgprot比较数值明显增高,差异有显著性意义(P<0.01)。[结论]紫锥菊中的多酚植物化学物质对大鼠应激性胃溃疡的预防性保护作用可能与其能提高机体免疫力和协调及增强NOS活力有关。[ Objective] To investigate the effect of polyphenol phytochemicals in echinacea on stress gastric ulcer in rats under restraint water -immersion stress. [ Methods] Fifty healthy SD rats (equal male and female) were randomly divided into five groups, which were control group with cimetidine, control group with normal saline (NS), rhodioloside group and echinacea extract group under restraint water stress. Ulcer index, spleen index, and nitric oxide synthase (NOS) activity of gastric mucosa were measured. [ Results] The ulcer index (UI) of echinacea extract group was (15 + 2) score, compared with UI (65 ±4) score of NS model group, there was a statistically significant decrease between them(P 〈0.01 ). The spleen index (SI) of echinacea extract group was (5. 425±0. 548), compared with SI (2.323 ±0. 392) of NS model group, there was a statistically significant increase between them ( P 〈 0.01 ). Otherwise, the NOS activity of gastric mucosa in glandular stomach and the body of stomach were (56. 249 ± 3. 473 ) U/mgprot and (53. 852 ± 6. 146 ) U/mgprot respectively, compared with NS model group (28.240±4.956) U/mgprot and (27. 173±4.244) U/mgprot, there was a statistically significant increase between them(P 〈0. 01 ). [ Conclusion ] Polyphenol phytochemicals in echinacea may prevent the gastric mueosal damage from stress in rats by improving their immunity, coordinating and increasing the activity of NOS.
