摘  要:目的:观察N-硝基L-精氨酸甲酯(L-NAME)投药大鼠的动脉内皮功能及结构改变并探讨其相互关系。方法:大鼠离体动脉环的张力测定,3氚-瓜氨酸标测及组织染色。结果:大鼠胸主动脉的内皮依赖性舒张功能在投药3天后明显下降,1周时有所恢复,8周时再次下降并释放内皮依赖性收缩因子。肠系膜微动脉的内皮功能无改变。投药8周后出现了动脉中膜肥厚及其周围纤维化。结论:L-NAME投药大鼠的胸主动脉内皮功能下降与投药时间呈非线性关系。肠系膜微动脉的内皮功能无改变。组织学变化与内皮功能变化之间无直接关系。? Objective:The changes and the relation between the function and structure of arterial endothelium were studied in rats treated with NnitroLarginine methyl ester(LNAME). Methods:The tension of arterial ring of thoracic aorta and mesenteric artery was determined as routine.The nitric oxide synthase(NOS) activity was measured by 3Hcitrulline assay and the histological staining was performed using hematoxylineosin and masson trichrome stainings. Results:The endotheliumdependent relaxation to acetylcholine(Ach) of the aorta was reduced 3 days,recovered at 1 week and again reduced at 8week after the treatment of LNAME;while the relaxation of small mesenteric arteries was unaltered throughout the experimental periods.At 8week treated group the indomethacinsensitive endotheliumdependent contractions to Ach and the medial thickening and perivascular fibrosis were observed. Conclusion:The endothelial function of rat aorta is decreased nonlinearly dependent on the administration time of LNAME,while that of small mesenteric arteries is unaltered throughout the experimental periods.There is on direct relationship between the histological variation and functional changes of endothelium.
