摘  要:目的探讨肾上腺切除SD大鼠过敏性哮喘模型的制备。方法切除SD大鼠肾上腺后,腹腔注射卵蛋白致敏,2wk后静脉内注射卵蛋白激发哮喘发作。结果卵蛋白激发后肾上腺切除大鼠气道阻力明显增加,最大增加时间较普通与假切组明显提前,升高幅度较普通组和假切组分别增加33.33%和77.78%。肾上腺切除大鼠卵蛋白激发后肺顺应性明显降低,与气道阻力的增加呈显著负相关关系。结论制备的肾上腺切除大鼠过敏性哮喘模型可用于非糖皮质激素类抗哮喘免疫抑制剂的研究,也可用于糖皮质激素抗哮喘作用原理的研究。Objective To prepare allergic asthma model of SD rat with adrenalectomy(ADX).Methods After ADX,SD rats were intraperitoneally injected with ovalbumin for immunization,and two weeks later the rats were intravenously injected with ovalbumin for attack. Results After attack with ovalbumin,airway resistance in ADX,normal and sham adrenalectomy rats were significantly increased,the peak of airway resistance in ADX rats were at 2 min after ovalbumin attack,but in normal and sham adrenalectomy rats were at 21 min after ovalbumin attack,the time of the peak of airway resistance between ADX and normal,sham adrenalectomy were significantly different. The amplitude of the peak of the airway resistance in ADX rats were higher than the amplitude in normal and sham adrenalectomy rats for 33.33% and 77.78% respectively.The lung compliance were significantly decreased in ADX rats after ovalbumin attack,and the decrement of the lung compliance were significantly correlated with the increment of the airway resistance in the ADX rats. Conclusions The preparation of the allergic asthma model in adrenalectomy SD rats were success,and the model was suitable to researches on nonglucocorticoids immunodepressive(s) for asthma therapy,and was also applicable to researches on the mechanisms of glucocorticoids treatment in bronchial asthma.
