摘  要:目的探讨姜黄素对体内人脑胶质瘤细胞U251生长的抑制作用。方法建立人脑胶质瘤细胞U251裸鼠皮下移植瘤模型后随机分为对照(C)组、姜黄素100mg/kg治疗(A)组和姜黄素50mg/kg治疗(B)组。腹腔注射给药30d,观察姜黄素治疗对移植瘤的体积抑制率和重量抑制率。治疗结束后收集各组皮下移植瘤组织,Westernblot法检测c-fos蛋白、细胞外调节蛋白激酶(ERK)、磷酸化细胞外调节蛋白激酶(p-ERK)表达。结果A、B两组移植瘤体积抑制率分别为62.4%、38.9%,重量抑制率分别为60.6%、36.4%。A、B两组肿瘤c-fos蛋白表达水平均明显低于C组,A组p-ERK蛋白水平明显低于C组。结论姜黄素对体内人脑胶质瘤细胞U251的生长有显著抑制作用。这可能与阻滞ERK信号通路有关。Objective To investigate the inhibitory effect of curcumin on the growth of human glioma U251 cells in vivo. Methods After establishing the nude mouse model of human glioma U251 cells by subcutaneous transplantation, the glioma-bearing mice were randomly assigned into 3 groups. The mice in group A and group B were intraperitoneally injected curcumin 100 mg/kg and 50 mg/kg, respectively,for a month. The inhibition rates of tumor volume and weight were calculated. The expressions of c-los, total and phosphorylated extracellular regulated kinase(tvERK) were assessed by Western blot analysis. Results The inhibition rates of tumor volume of group A and B were 62.4 and 38. 9% ,and those of weight were 60.6% and 36.4%, respectively. The expression of c-fos was lower in group A and 13 than that in group C. The expression of p-ERK was lower in group A than that in group C. Conclusion Curcumin has a significant inhibitory effect on the growth of U251 cells in vivo,which may be related to its blocking effect on the ERK pathway.
