摘  要:T细胞在机体应对外来抗原的免疫应答过程中处于其中心环节。木犀草素(luteolin)有抗炎、抗氧化、抗肿瘤、抗变应性、抗病毒和细胞保护作用。研究中,以佛波醇酯(PMA)+离子霉(Ion)素刺激小鼠淋巴细胞来源的T细胞,以不同终浓度的木犀草素与T细胞共培养,MTT法检测发现,PMA+Ion刺激48 h后,小鼠T淋巴细胞增殖显著高于对照组;木犀草素(10、20、30μmol/L)可显著降低PMA+Lon引起的细胞增殖;Western blot检测发现,PMA刺激T细胞30 min时能显著促进胞浆中的p65进入核,木犀草素(10、20、30μmol/L)能抑制PMA刺激的T细胞p65入核,以30μmol/L的木犀草素抑制作用最强。研究表明,木犀草素能通过抑制NF-κB p65的入核,抑制小鼠T细胞的增殖和活化,调节机体免疫功能。Luteolin processes various biological activities, such as anti-inflarnmatory effect, anti-oxidation effect, anti-aller- gy effect, anti-virus effect, anti-tumor effect and cell-protection effect. In the present study, T cells from mouse were stim- ulated by phorbol 12-myristate 13-acetate (PMA) plus ionomycin (Ion) and then followed by stimulation with different concen- trations of luteolin. By using MTT assay, it was found that T cells markedly proliferated after stimulation with PMA plus Ion for 48 hours in comparison with the control group. Luteolin in concentrations of 10, 20 and 30μmol/L could decade the T cell proliferation induced by stimulation with PMA plus Ion. As demonstrated by Western blot assay, the migration of p65 protein from cytoplasm into nucleus was significantly enhanced when T cells were stimulated with PMA plus Ion for 30 minutes and this effect could be inhibited by luteolin at dose of 10, 20 and 30 μmol/L. The most prominent effect appeared with ao 30μmol/ L of luteolin. These results indicate that luteolin can prevent the activation and proliferation of murine T cells, probably due to inhibition of migration of p65 protein from cytoplasm into nucleus in the activated T cells.
