摘　　要：研究单纯疱疹病毒侵入介质(herpesvirus entry mediator,HVEM)在人CD4^+CD25^+调节性T细胞(Treg)上的表达及对Treg功能的影响。分离人外周血单个核细胞,磁珠纯化获得T细胞及其不同亚群,流式细胞术检测Treg上HVEM的表达。Treg按不同比例分别与去除Treg的CD3^+T细胞、CD4^+CD25^-T细胞或CD8^+T细胞混合培养,加入抗HVEM抗体或同型对照抗体,并用CD3抗体单独或联合CD28抗体刺激T细胞,MTT法检测T细胞增殖。结果显示,Treg上HVEM表达量为90%～95%(n=15)。抗HVEM抗体组与对照组相比,降低了Treg对各类T细胞增殖的抑制作用(P<0.05),证明Treg表达的HVEM介导了其发挥抑制功能。Analysis of the expression of herpes virus entry mediator （HVEM） on human CD4＋ CD25＋ regulatory T cells （Treg） and the study of its effect on Treg function were investigated in the present study. After separation of peripheral blood mononuclear cells, total and subset T cells were selected by using magnetic beads, and the expression of HVEM on CD4＋ CD25＋ T cells was detected by flow cytometry. The CD3＋ T cells without inclusion of Treg cells, CD4＋ CD25 T cells and CDS＋T cells were co-cultured with different ratios of Treg respectively, in the presence of anti-HVEM antibodies or isotype of IgG. Proliferation of T cells stimulated by CD3 mAb or combined with CD28 mAb was measured by MTT assay. The resuits showed that Treg constitutively expressed high level of HVEM, ranged from 90% to 95% （n=15）. Blockade of HVEM on Treg significantly attenuated the inhibition of Treg on different sorts of T cells compared with isotype IgG group （P〈 0.05）. These results demonstrate that HVEM mediates the inhibitory function of Treg cells＇ inhibitory function.
