摘 要:目的:观察小檗碱对体外血管平滑肌细胞(VSMCs)增殖、迁移和凋亡的影响。方法:培养兔主动脉平滑肌细胞至8~10代,根据小檗碱作用时间不同分成24h、72h组,每一时间组据小檗碱浓度不同分为5组:0μmol/L、25μmol/L、50μmol/L、100μmol/L和200μmol/L组。用台盼蓝活细胞计数法和四甲基偶氮唑蓝(MTT)法观察药物对VSMCs增殖的影响。用Boyden小室法检测穿膜细胞数,观察小檗碱对VSMCs迁移的影响。用TUNEL法及流式细胞仪检测小檗碱对VSMCs凋亡的影响。结果:台盼蓝活细胞计数显示小檗碱作用24h、72h后,随小檗碱浓度增加,活细胞数减少(P<0.05)。MTT法检测显示小檗碱作用24h、72h后,随小檗碱浓度增加,生长抑制率增高,随小檗碱干预时间延长,细胞生长抑制率增加(P<0.01)。Boyden小室法检测显示随小檗碱浓度增加,穿膜细胞数减少(P<0.05)。TUNEL检测显示各组施加干预48h后,对照组和各实验组的凋亡阳性率分别为13.15%、12.04%、10.62%、9.73%和9.72%(P>0.05)。流式细胞仪检测小檗碱作用48h后,对照组、50μmol/L和100μmol/L浓度小檗碱组的细胞凋亡率分别为20.4%、18.7%和17.2%。结论:小檗碱对VSMCs的增殖和迁移有显著抑制作用,而小檗碱对VSMCs的凋亡无促进作用。Objective: To investigate the effects of berberine on the preliferation, migration and apoptosis of vascular smooth muscle cells (VSMCs) in vitro. Methods: We cultivated the VSMCs. They grew from 8th to llth generation. The VSMCs were devided into two groups: the 24 hours group and the 72 hours group. And there were four different groups in every time group according to various concentrations of berberine: 0μmol/L, 25μmol/L, 50μmol/L, 100μmol/L and 200μmol/L. Cell proliferation was determined by direct cell counting with trypan blue and methylthiazoletrazolium staining. We used Boyden chamber to count the amount of migratory cells to detect effects of berberine on the migration of VSMCs. We detected the effect of berberine on the apoptosis of VSMCs by the terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL) and the flow-cytometer (FCM) technique. Results: The quantites of cells of unique density of berberine group after 24 hours and 72 hours decreased,compare to vs control group with trypan blue staining(P 〈 0. 05). With the increasement of the concentration of berberine, the amount of ceils decreased. With the increase of the concentration of berberine, the amount of cells that cutting through the cellulose nitrate membranes (CN) decreased. The rate of TUNEL positive cells dealed with 0μmol/L, 50μmol/L, 100μmol/L berberine for 48 hours was 13.15%,12.04%,10. 62%,9.73% and 9.72%(P〉0.05). The apoptosis ratio of rate of 0μmol/L group, 50μmol/L group and 100μmol/L group dealed with berberine after 48 hours was 20. 4 M,18.7 %and 17.2 %. Conclusion.. Berberine could inhibit the proliferation and migration of VSMCs. But it does not induce the apoptosis of VSMCs.
