摘 要:目的观察通心络对链脲佐菌素(STZ)糖尿病大鼠心肌微血管炎症损伤及血管痉挛的防治作用。方法38只STZ糖尿病大鼠分为STZ对照组、胰岛素组、通心络组,另取8只正常Wistar大鼠作为正常对照组。正常对照组、STZ对照组予生理盐水灌胃,胰岛素组皮下注射胰岛素,通心络组灌胃通心络混悬液,共8周。取心肌切片作HE染色、组织核因子-κB(NF-κB)免疫组化染色。取心肌匀浆测定细胞间粘附分子-1(sICAM-1)、肿瘤坏死因子α(TNF-α)、内皮素-1(ET-1)、血管紧张素-Ⅱ(AT-Ⅱ),取血浆测定AT-Ⅱ、神经肽Y(NPY)。sICAM-1采用酶联免疫法,其余采用放射免疫法。对NF-κB染色切片采用计算机图像分析,取其阳性面密度比值的均值以评价其表达程度。结果与正常大鼠比较,STZ糖尿病大鼠心肌匀浆sICAM-1、TNF-α、ET-1、AT-Ⅱ含量及NF-κB阳性表达升高(P<0.01或0.05),通心络组降低(P<0.01或0.05),胰岛素组有升高或降低趋势(P<0.05或P>0.05)。STZ糖尿病大鼠血浆AT-Ⅱ升高(P<0.05),通心络组AT-Ⅱ升高(P<0.05)。STZ糖尿病大鼠血浆NPY明显降低(P<0.01),通心络组NPY升高(P<0.01),而胰岛素组明显降低(P<0.01)。通心络组血糖无明显变化。HE染色显示通心络组心肌病变减轻。结论通心络对STZ糖尿病大鼠心肌微血管炎症损伤及血管痉挛具有防治作用。Objective To observe the preventative and curative effect of Tongxinluo on microvascular inflammatory injury and vasospasm in the cardiac muscle of streptozotocin (STZ) diabetic rats. Methods STZ rats were divided into STZ group, insulin group, Tongxinluo group, with the other 8 Wistar rats as normal control. The normal group and STZ group were intragastric administered with NS, insulin group was subcutaneous injected with insulin, and Tongxinluo group was intragastric administered with Tongxinluo suspension for 8 weeks. The sections of cardiac muscle was stained in HE staining and NF- κ B immunohistochemical staining. The level of adhesion molecule-1 (sICAM-1), tumor necrosis factor (TNF- a ),endothelin-1 (ET-I), angiotensin-II (AT-II) in myocardial homogenate from intercellular, and level of AT-II, neuropeptide Y (NPY) in plasma were determined, with sICAM- 1 by enzyme-linked immunosorbent assay and the others by radioimmunoassay. The sections stained by NF-κ B immunohistochemical staining were image analyzed using computer. The extent of its expression was evaluated by the mean of the density ratio of the positive side. Results Compared with normal rats, the content of sICAM-1, TNF-a, ET-1, AT-II and the expression of NF- κB in the myocardial homogenate of STZ rats was increased (P〈0.01-0.05),while that in Tongxinluo group was reduced (P 〈0.01-0.05), and they had the trend of increase or decrease in the insulin group (P〈0.05 or P〉0.05). AT-II in plasma of STZ rats increased (P〈0.05), but in the Tongxinluo group it was increased (P〈0.05). The level of NPY in STZ rats plasma was significantly reduced (P〈0.01), but it was increased in the Tongxinluo group (P〈0.01), and in the insulin group it was significantly reduced (P〈0.01). Blood glucose had no significant change in Tongxinluo group. HE staining showed the pathological changes of cardiac lesion in Tongxinluo group was significantly mitigated.
