双单链抗体夹心法诊断汉坦病毒感染的初步研究

摘　　要：目的制备检测汉坦病毒感染的双单链抗体夹心ELISA诊断试剂,并予以评价。方法以抗汉坦病毒NP抗原单链抗体包被反应板,以辣根过氧化物酶(HRP)标记大肠埃希菌表达的抗NP抗原单链抗体,并制成双单链抗体夹心诊断试剂,检测56份早期肾综合征出血热(HFRS)患者血清及66份非HFRS对照血清。结果56份HFRS患者血清中,检测出29例阳性,阳性率为51.8%,对照组66例,检测结果均为阴性。结论研制的试剂特异性好,价格低廉。Objective A double scFv sandwich ELISA diagnosis reagent was prepared to detect Hantavirus infection. Methods Single -chain fragment of variable region（ scFv）, which was expressed in E. coli, against nucleocapsid protein of Hantavirus （NP） antigen was labeled with Peroxidase horseradish（HRP） and ELISA plates were coated with scFv against NP antigen to prepare a double scFv sand- wich diagnosis reagent to detect 56 sera of early stage HFRS patients and 66 sera in control group. Results 56 sera, of which 29 sera were positive, of HFRS patients ：were detected by the double scFv sandwich and positive rate was 51.8%. 66 sera in control group were all negative. Conclusion The prepared double scFv sandwich reagent was specific and cheap.
