摘 要:目的:观察麝香酮对易卒中型肾血管性高血压大鼠(RHRSP)血浆内皮素(endothelin,ET)和脑皮质微血管结构改变的影响。方法:复制肾血管性高血压大鼠动物模型,将雄性RHRSP自术后第8周分成两组:高血压组和麝香酮治疗组,将年龄、性别、数量相同的假手术组作对照组,测量收缩压(SBP)和12周时血浆ET含量,光镜和电镜下观察脑皮质微血管形态改变。结果:12周时麝香酮组的SBP水平比高血压组低[(23.95±0.73)kPa vs(26.13±2.22)kPa,P<0.05],ET水平也比高血压组低[(73.25±10.78)pg/mLvs(89.44±9.92)pg/mL,P<0.05]。光镜和电镜下观察均发现高血压组皮质微血管损害最严重,麝香酮组微血管形态损害比高血压组轻。结论:麝香酮可降低RHRSP的SBP和血浆ET水平,并能在一定程度上改善肾血管性高血压大鼠脑皮质微血管损害。Objective:To observe the effects of Muscone on the plasma endothelin and structural changes of cerebral cortex microvessels of stroke-prone renal vascular hypertensive rats (RHRSP). Methods: The animal model of RHRSP was divided into two groups at 9 weeks after operation, the hypertension and Muscone groups. The same age, gender, amount of sham-operated rats was used as normotensive control group. The systolic blood pressure (SBP) and the level of plasma endothelin at 12 weeks were measured in each group, and the structure of cerebral cortex microvessel was observed under light microscope and electron microscope at 12 weeks after operation. Results:The SBP( 23.95 ± 0. 73) and the level of plasma endothelin(73.25 ±10.78 pg/ml) of Muscone group at 12 weeks after operation were lower than hypertension group( sbp: 26.13 ±2.22 ,endothelin: 89.44 ±9.92 pg/mL) (P 〈0.05). The Morphological changes of the cerebral eortex microvessels of hypertension group were most severely impaired, and the Museone group was not so severely impaired than those of hypertension group. Conclusion:The Museone treatment can decrease the SBP and the plasma endothelin level of RHRSP, and effectively improve its impaired microvessel of hypertensive cerebral cortex on some degree.
