p21与TGF-β在大鼠肾间质纤维化模型中的表达

摘　　要：目的:探讨p21及TGF-β在肾间质纤维化中的作用及其可能的调控机制。方法:将SD大鼠随机分为假手术组(SOR组)和单侧输尿管结扎组(UUO组)。HE染色观察2组大鼠术后第7,14和21天肾小管间质的形态学改变;用RT-PCR法检测肾皮质p21 mRNA和TGF-βmRNA的变化。结果:在UUO大鼠模型早期,肾皮质p21 mRNA和TGF-βmRNA的表达即有明显增高,与SOR组大鼠比较,差异有显著性(分别P<0.01及P<0.05),随着梗阻时间的延长,p21和TGF-β的表达逐渐增强(P<0.05),并且UUO组大鼠各时间点p21和TGF-β的表达呈正相关(各时间点分别为r=0.745、0.700、0.662,P<0.05)。结论:p21可能参与了单侧输尿管梗阻大鼠肾小管间质纤维化的发病过程,TGF-β可能参与介导了p21在肾皮质表达的调控。Objective：To observe the expression of p21 and TGF-βin renal interstitial fibrosis rats,investigate the effect of p21 expression and the regulation mechanisms of it. Methods： Sprague-Dawley rats were randomly divided into shame operation rats group and unilateral urethral obstruction. Histological change were observed by HE stain in different times（7,14,21 after UUO, sham-surgery or enalapril treatment）; The expression of p21 mRNA and TGF-βmRNA was detected by RT-PCR. Results： Seven days surgery, significant differences were found in p21 and TGF-β expression between UUO and SOR renal tubular cells. With degree of interstitial fibrosis aggravating, the expression of p21 and TGF-β increasing. Expression of p21 was positive correlation with TGF-β level（ r =0. 745,0. 700,0. 662 ,P 〈 0.05）. Conclusion： p21 may participate in the pathogenesis of renal tubule-interstitial fibrosis. TGF-β expression increasing in renal tubular epithelial cell play a role in the progression of renal interstitial. Key words p21 ;TGF-β;renal; fibrosis
