摘 要:目的研究卡巴胆碱(CAR)对脓毒症大鼠肝组织缺血引起的脂质过氧化损伤的保护作用。方法雄性SD大鼠32只,采用盲肠结扎穿孔术(CLP)制作大鼠脓毒症模型。随机分为CLP组、卡巴胆碱干预组(CAR组),每组16只。CLP术后立即静脉注射CAR10μg/ks(CAR组)或等量生理盐水(CLP组)。采用激光多普勒血流仪测定大鼠CLP术后6h和12h肝脏血流量;然后处死动物,取肝组织,测定丙二醛(MDA)含量、黄嘌呤氧化酶(XOD)和二胺氧化酶(DAO)活性及组织含水量。结果CLP术后6、12hCAR组肝脏血流量高于CLP组(P〈0.05)。CAR组肝脏XOD活性、MDA含量和ALT水平显著低于CLP组(P〈0.05),6h组、12h组差异均有统计学意义。CAR组肝脏组织含水量均低于CLP组(P〈0.05),6h组、12h组比较差异有统计学意义(P〈0.05)。结论卡巴胆碱能改善脓毒症大鼠肝缺血,抑制氧自由基生成,减轻肝组织水肿和功能损害。Objective To investigate protective effect of carbachol on on liver free radical injury in rats with sepsis. Methods Thiyty two Sprague - Dawley rats were projected to sepsis induced by cecal ligation and puncture( CLP) , and divided randomly into two equal groups: septic model group( CLP group) and septic model with CAR- treatment group( CAR group). Mter operation, CAR 10 μg/kg (CAR group) or saline (shame and CLP group) was immediately injected into penis vein. The blood flow of liver was detected by a laster - doppler flowmetry during the experiment. Animals were sacrificed at 6 hour and 12 hour after CLP respectively, and sepcimens of liver were harvested for evaluation of xanthine oxidase( XOD ), malondialdehyde( MDA ) and assessment of the rate of water content (ratio of dry and wet weight ) of liver. Results The blood flow of liver of CAR group were significantly higher than those of CLP group both 6 h and 12 h,repectively (6 h,68. 92 ± 10. 45 VS 42. 61 ± 10. 37,P 〈 0. 05 ; 12 h,42.49 ± 9. 54 VS. 30. 12 ± 8.27 ,P 〈 0. 05 ). It was showed signifant lower levels of XOD, MDA and ALT in CAR group than CLP group in liver both 6 h and 12 h, repeetively ( P 〈 0. 05 ). The rates of tissue water contents of CAR group were obviously lower than those of CLP group in liver both 6 h and 12 h,repectively{ 6 h,70. 9 ±2.9 VS. 75.2 ±2. 2,P 〈0. 05;12 h,71.6 ±2. 7 VS. 74. 8 ±2. 2, P 〈0. 05). Conclusion The results indicate that carbachal exert its protective effects by promoting liver peffusion, inhibiting lipid oxidative prouction and alleviate liver edema and dysfunction in rates with sepsis.
