摘 要:将荷人B细胞淋巴瘤裸鼠分为对照组、Fab抗体组、单次注射组和重复注射组,尾静脉注射131I-Fab抗体后进行ECT显像;每隔5d用MRI结合显微镜线圈检测荷瘤裸鼠的肿瘤大小。注射后25d,取出肿瘤,进行病理分析、流式细胞术和电镜检测细胞凋亡。结果显示,131I-Fab抗体静脉注射后8h在肿瘤局部浓集明显,瘤体清晰显像;Fab组5d内具有抑制肿瘤生长作用,凋亡率为(2.48±0.6)%;单次131I-Fab注射组早期抑制肿瘤生长较明显,肿瘤生长出现平台期,凋亡率为(10.67±1.6)%;131I-Fab抗体重复注射组抑制肿瘤生长作用明显,肿瘤局部坏死脱落,凋亡率为(14.00±1.9)%。病理结果显示两注射组均可见组织坏死。MRI图像与病理结果相符,通过造影剂增强可提供较准确的病理信息。表明131I-Fab抗体具有早期ECT肿瘤显像、瘤体准确定位,并通过致凋亡和直接促使细胞死亡的作用而有效抑制肿瘤生长,大型高场强MRI可为小动物模型实验提供有效、无创、动态的监测。Nude mice bearing human B cell lymphoma were divided into four groups: control, Fab antibody, single injection and duplication injection. Radioimmunoimages were taken after injection with ^131I-labeled Fab antibody through tail vein. The cancer-bearing mice were detected by MRI combined with microscopy coil every 5 days, and tumor sizes were measured. On the 25th day after injection, the tumors were taken out for pathology and apoptosis analysis by flow cytometry (FCM) and TEM, respectively. The results of ECT showed that radioactive nuclide enriched in the area of the tumor obviously at 8 hour after injection, and the mass could be displayed clearly. The growth suppression effect could be displayed in 5 days in Fab antibody group and the apoptotic rate was (2.48±0.6)%. The growth suppression effect was obvious in the prophase in single injection group, and the plateau phase could be seen in the tumor growth curve, with the apoptotic rate of (10.67±1.6)%. The growth suppression effect was more obvious in duplication injection group and the local tissues of tumor necrosed and fallen off, with the apoptotic rate of (14.00±1.9)%. The pathologic results showed tissue necroses in both single group and duplication injection group. The MRI image conformed to the results of pathology, and could provide accurate pathology information by contrast medium enhancement. ^131I labeled Fab antibody against B cell lymphoma can image the tumor early through ECT, locate the tumor mass accurately and suppress tumor growth through the mechanism of apoptosis and cell death. It can be a kind of potential and effective bioengineering medicine to treat the tumor in the area of radioimmunotherapy. Large high field MR/can provide effective, atraumatic and dynamic detection to the little animal model, and have wide application for little animal model.
