摘 要:目的探讨姜黄素对脑缺血模型大鼠SOD酶活性及Akt磷酸化水平的影响。方法48只SD大鼠随机均分为假手术组、手术模型组、姜黄素低、高剂量组。采用Zea Longa线栓法制作模型,检测各组大鼠的SOD活性和Akt磷酸化水平。结果与假手术组比,模型组和姜黄素低、高剂量组SOD活性下降;与模型组比较,姜黄素低、高剂量组SOD活性升高(P<0.05),但姜黄素低、高剂量组的SOD活性差异无显著性(P>0.05);与假手术组比较,模型组的Akt磷酸化水平显著下降(P<0.01),姜黄素低、高剂量组Akt磷酸化较模型组显著升高(P<0.01),且高剂量组的Akt磷酸化水平较低剂量组高(P<0.01)。结论姜黄素对脑缺血的保护作用可能与提高SOD活性及Akt磷酸化水平有关。OBJECTIVE To investigate the underlying mechanism of neuroprotective effect of curcumin against focal cerebral ischemia in rat.METHOD Forty-eight SD rats were randomly divided into 4 groups:sham operation group,middle cerebral occlusion(MCAO) group,low dose curcumin group and high dose curcumin group.Middle cerebral occlusion was induced by means of a intraluminal nylon suture.The activity of SOD and the phosphorylation level of Akt were tested in each group.RESULT The SOD activity was decreased in MCAO group and curcunin compared with sham group(P〈0.05).An increased of SOD activity was observed in low and high dose curcumin groups compared with MCAO group(P〈0.05),but there is no difference between low and high dose curcumin group(P〉0.05).The level of phospho-Akt was significantly decreased in MCAO group compared with sham group(P〈0.01).Compared with MCAO group,the level of phospho-Akt was significantly increased in low and high dose curcumin groups(P〈0.01),and a higher level of phospho-Akt was observed in high dose curcumin group compared with low dose curcumin group(P〈0.01).CONCLUSION The underlying mechanism of neuroprotective effect of curcumin against focal cerebral ischemia in rat may be involved in increased SOD activity and upregulating phospho-Akt level.
