摘 要:分别克隆番茄PR-1和PR-5基因片段,并以之制备探针,采用Northern blot方法研究了PR-1和PR-5基因在正常生长条件下番茄根、茎、叶、果实发育中的表达模式,研究了干旱、复水和内外源乙烯对这两种基因表达的影响.结果表明:这两个基因主要在衰老叶片、B期果实和根部表达,干旱抑制了这两个基因的表达,复水和乙烯诱导其表达.PR-1 and PR-5 gene fragments were isolated with PCR and RT-PCR from the genomic DNA and eDNA of tomato leaves. Then the specific DNA fragments of PR-1 and PR-5 genes were used as probes to hybridize with the total RNAs extracted from the wild type tomato (AC^++) and the ethylene response mutants of tomato (Nr, T4B-11) to analyse the expression pattern of the two genes in tomato root, stem, leaf and fruit in normal conditions and to study the effect of drought, re-watering and exogenous and endogenous ethylene on the expression of the two genes. Northern blot hybridization results indicated that PR-1 and PR-5 genes were expressed mainly in senescent leaves, fruit at Stage B and roots,and thheir expression was restrained by drought and induced by re-watering and ethylene.
