摘 要:目的探讨柯里拉京对沙土鼠脑缺血再灌注损伤的保护作用。方法沙土鼠60只,雌雄各半,随机分成6组,每组10只,即假手术组、模型组、柯里拉京5 mg/kg组、10 mg/kg组、20 mg/kg组(柯里拉京5 mg组,柯里拉京10 mg组,柯里拉京20 mg组)和尼莫地平1 mg/kg组(尼莫地平1 mg组)。采用夹闭沙土鼠双侧颈总动脉10 min,继以再灌注5天建立沙土鼠脑缺血再灌注损伤模型,评价柯里拉京对沙土鼠脑电图及大脑皮质水、Ca^(2+)、Na^(+)含量的影响。结果再灌注损伤后,模型组脑电图电位幅度持续低平,与假手术组比较差异有统计学意义(P<0.01)。柯里拉京10 mg组、20 mg组可促进脑电图电位幅度的恢复,与模型组比较有显著差异(P<0.01);再灌注损伤后5天,柯里拉京10 mg组、20 mg组可降低大脑皮质水和Ca^(2+)含量,与模型组比较有显著差异(P<0.05,P<0.01),但Na^+含量无显著差异(P>0.05)。结论柯里拉京对沙土鼠脑缺血再灌注损伤具有较好的保护作用,其机制与降低大脑皮质含水量及Ca^(2+)含量密切相关。Objective To study the protective effects of corilagin against cerebral ischemia-reperfusion injury. Method Sixty gerbils were randomly divided into six groups(5 male and 5 female gerbils in each group) :sham operation control group, model group, three corilagin groups(5 mg/ kg,10 mg/kg and 20 mg/kg) and nimodipine group. The model of cerebral ischemia-reperfusion injury was produced by 10-min occlusion of bilateral common carotid arteries followed by 5-day reperfusion. The EEG was recorded after reperfusion. Contents of water, calcium and sodium in cerebral cortex were determined after 5-day reperfusion. Results Corilagin groups(10 mg/kg and 20 mg/kg) significantly improved the recovery of electroencephalogram potential amplitude after reperfusion, which was significantly higher than that in model group (P 〈 0.01). Corilagin groups (10 mg/kg and 20 mg/kg) significantly decreased contents of water and calcium in cortex after 5-day reperfusion,which were significantly lower than those in model group (P 〈 0. 05, P〈 0.01). However,no significant effect on cortex sodium content was observed (P〉 0.05). Conclusion Corilagin showed protective effects against cerebral ischemia-reperfusion injury in gerbils, and its mechanism might be related to reducing calcium and water contents in cerebral cortex.
