石榴皮对实验性胃损伤及相关机制的研究

摘　　要：目的:研究石榴皮丙酮提取物对实验性胃损伤的防治作用及其与胃粘膜前列腺素E2的关系。方法:采用大鼠幽门结扎型胃溃疡模型和乙醇诱导的胃粘膜损伤模型,观察石榴皮丙酮提取物灌胃对实验性胃损伤及胃液、胃酸、胃黏液和胃蛋白酶分泌的影响;采用放射免疫方法研究了石榴皮提取物对幽门结扎所致胃损伤大鼠胃粘膜前列腺素E(PGE2)含量的影响。结果:石榴皮提取物(50mg/kg,150mg/kg和50mg/kg)灌胃呈剂量依赖性明显抑制大鼠幽门结扎型胃溃疡的形成和无水乙醇致胃粘膜损伤的发生,促进胃黏液的分泌且能增加PGE2水平,但对胃液分泌、游离酸度、总酸度和总酸排出量及胃蛋白酶活性无明显影响。结论:石榴皮提取物对实验性胃溃疡具有良好的防治疗作用,这种作用可能与促进胃黏液分泌,维护粘膜屏障的完整性和增加胃粘膜PGE2合成有关。To observe the effects of acetone extract obtained from pomegranate bark on the experimental gastric damages and possible mechanism involved in the levels of PGE2 in gastric mucosa. Methods ： Gastric damage models were established by pylorus ligation and intragastric absolute ethanol, respectively. The ulcer index, the gastric juice volume, free acidity, total acidity, total acid output, the pepsin activity, the amount of adherent mucus and free mucus , as well as the levels of PGE2 in mucosa were measured, respectively. Results：The acetone extract of pomegranate bark（50 mg/kg, 150 mg/kg, and 500 mg/kg） significantly inhibited ulcerative formation induced by pylorus ligation, obviously decreased the gastric mucosa damages induced by intragastrie absolute ethanol, in dose-dependent manner. It also significandy increased the levels of PGE2 in mucosa and promoted the secretion of adherent mucus and free mucus without influence on the elevations of free acidity, total acidity, total acid output, gastric juice volume, and gastric pepsin activity induced by pylorus ligation. Conclusion： The acetone extract of pomegranate bark play a protective role against gastric damages, its the mechanism may be related to increasing the levels of mucous PGE2, secretion of adherent mucus and free mucus from the stomach wall.
