摘  要:目的:建立并验证银杏叶提取物(GBE)血清药理学研究方法。方法:MTT法确定体系中血清添加量;HPLC法结合药效法确定大鼠给药方案及采血时间;以T-SOD、MDA为指标测定该含药血清的抗氧化能力。结果:大鼠按人临床用药等效剂量的5倍,每日两次连续4天灌胃给予GBE,于末次给药后2h采血时血清中GBE各有效成分的含量最高;体系中加入20%该条件下制备的含药血清对大鼠肾小球系膜细胞(MC)正常增殖没有影响;采用该方法制备的GBE含药血清具有明显的抗氧化能力。结论:本实验条件下制备的GBE含药血清具有明显的抗氧化能力,该血清药理学方法能用于GBE含药血清药效及机制研究。To establish and validated a serum pharmacology method for Ginkgo biloba extract (GBE). Methods: MTT assay was used to determine the suitable concentration of blank serum in culture system. HPLC method was use to determine the optimum dosage of administration and time of hemospasia. T-SOD and MDA was selected to validate the antioxidant effect of serum containing GBE. Results: Rats were oral administration of GBE by 5 times dose of adults twice a day, keeping on 4 days. Hemospasia after 2 hours of the last administration was the best preparative method of the serum containing drug. Adding 20% of this serum to culture system has no influence on multiplication of mesangial cells (MC). This serum containing GBE has obvious antioxidant effect on MC. Conclusion: Serum containing GBE prepared by suitable method possesses obvious antioxidant effect and this serum pharmacology method can be applied to evaluate the pharmacological effect and mechanism of GBE.
