摘  要:目的:探讨苦参碱(MT)预处理对大鼠肝脏缺血再灌注损伤的影响。方法:50只雄性SD大鼠随机分为5组:假手术组(SO组)、缺血再灌注组(NS组)和不同剂量MT(3.75、7.5、15mg/kg)预处理组(MT1、2、3组)。动物麻醉后分别经门静脉注入生理盐水或MT,然后阻断左、中叶肝蒂45min,造成70%肝脏缺血,SO组仅行分离不阻断,再恢复供血40min。术后采集下腔静脉血测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)水平,取肝组织测定超氧化物歧化酶(SOD)与丙二醛(MDA)水平,并行肝组织病理学观察。结果:MT预处理组血清ALT、AST、LDH水平及肝组织MDA含量低于NS组,肝组织SOD含量则高于NS组;MT预处理组肝细胞形态学异常改变较NS组明显减轻。结论:MT预处理可减轻大鼠肝脏缺血再灌注损伤,其机制可能与提高肝组织SOD含量,抑制脂质过氧化,保护线粒体有关。Objective: To investigate the protective effects of matrine (MT) preconditioning a- gainst hepatic ischemia-reperfusion injury in rats. Methods: Fifty male SD rats were randomly allocated into 5 groups: sham operation(SO) group, ischemia-reperfusion (NS) group and MT precondi- tioning groups (of 3.75, 7.5, 15 mg/kg). Rats of NS and MT groups were injected normal saline or MT, respectively; and 15 min later, the pedioales of the left and median liver lobes were clampped for 45 minutes, and reperfused for 40 min. The portal trial were only separated but not obstructed in SO rats. After reperfusion, the serum activity of ALT, AST and LDH was determined, the activity of SOD and content of MDA in liver tissue were examined, and histopathologica/changes of liver were observed. Results:Compared with those of NS group, the serum levels of ALT, AST, LDH. and the content of MDA in liver tissue of MT groups were lower, and the activity of SOD was higher(P〈0.05), The abnormal changes of the morphology of hepatocyte in the MT groups were improved. Conclusion: Precondition with MT can increase the activity of SOD in the hepatic tissue, inhibit lipid peroxidation and protect mitochondria; which could lead to reducing the liver injury caused by ischemia-reperfusion.
