摘  要:建立蒙古绵羊输卵管上皮细胞培养体系作为体外试验模型,根据体内雌激素浓度确定17-β-雌二醇添加的浓度分别为10-8、10-9、10-10、10-11mol/L,提取细胞总RNA,利用实时荧光定量RT-PCR测定sBD-1-mRNA的相对表达量。结果显示:一定浓度范围内(10-8、10-9、10-10mol/L),17-β-雌二醇对培养的输卵管上皮细胞sBD-1-mRNA的表达有促进作用,且呈正相关。结果表明:雌性生理周期下,雌性生殖道sBD-1-mRNA的表达与雌激素相关。To confirm the relationship between 17-β-estradiol and the expression of sBD-1 mRNA in female reproductive tract of Mongolian sheep, the epithelial cell of the fallopian tubes of sheep was established as in vitro model. Based on the concentration of estrogen of sheep in vivo, different concentrations of 17-β-estradiol (10^-8, 10^-9 , 10^-10 and 10^-11 mol/L) were added, following by extraction of the total RNA from cells, and the relative ex- pressions of sBD-1 mRNA were obtained using real time RT-PCR. The results showed that 17-β-estradiol had a positive impact on promoting the relative expression of sBD-1 mRNA in the epithelial cell of the fallopian tubes within the indicated concentrations (10^- 8, 10^-9 and 10 ^-10 mol/L). These results illuminated the expression of β-defensin was related to estrogen in female reproductive tract.
