摘 要:以新牧1号杂花苜蓿和新疆大叶紫花苜蓿的子叶作为外植体,研究激素配比、培养基种类、添加物等对苜蓿愈伤组织诱导和分化的影响。结果表明:新牧1号杂花苜蓿愈伤组织诱导的较优激素配比是2.00 mg/L2,4-D+0.05 mg/L KT,其诱导率达100.0%,三周愈伤直径大小达6.10 mm;较优分化培养基种类是SH培养基,分化率可达58.3%;在MS培养基中添加低浓度KT可使新牧1号杂花苜蓿获得较优的分化效果。新疆大叶紫花苜蓿较优分化培养基种类是SH培养基,分化率可达54.2%,以MSO为基本培养基,添加2.00 mg/LAgNO_3,可使新疆大叶杂花苜蓿的分化率从41.1%增加到63.3%。The experiment was designed to study the effect of hormone combination, medium types, and additives concentration on alfalfa callus induction and differentiation with cotyledon of Medicago varia Martyn.cv. Xinmu No. 1 and Medicago sativa L. cv. Daye as explants. The results showed that the better hormone combination for callus induction of Medicago var/a Martyn. cv. Xinmu No. 1 was 2.00 mg/L 2, 4 - D + 0. 05 mg/L KT, the induction rate was 100 percent, three weeks later, the callus diameter reached 6.10 mm; the optimum medium for differentiation was SH medium, its differentiation rate was 58.3%, the optimum hormone combination for differentiation of Medicago. var/a Martyn. cv. Xinmu No. 1 was MS added with low concentration KT. As for Medicago sativa L. cv. Daye, the optimum medium for differentiation also was SH medium, the differentiation rate was 54.2%, with MSO as the basal medium, adding 2.00 mg/L AgNO3 could increase differentiation rate of Medicago sativa L. cv. Daye from 41.1% to 63.3%.
