摘 要:目的:探讨中药单体榄香烯(effects of elemene,Ele)对人晶状体上皮细胞系(human lens epithelial cells B3,HLE-B3)内Ⅰ型胶原蛋白和Ⅲ型胶原蛋白合成的影响。方法:利用10μg/L碱性成纤维细胞生长因子(recombinant human basic fibroblast growth factor,rhbFGF)诱导HLE-B3增殖,将80mg/L的Ele作用在处于增殖状态下的HLE-B3,24h后采用双抗体夹心ABC-酶联免疫吸附测定法(enzyme-linked immunosorbent assay,ELISA)检测Ele作用后HLE-B3内Ⅰ型胶原蛋白和Ⅲ型胶原蛋白表达。结果:rhbFGF作用后HLE-B3内Ⅰ型胶原蛋白浓度为53.5±5.4μg/L,较正常组(38.5±2.3μg/L)明显升高,Ele作用后HLE-B3内Ⅰ型胶原蛋白的浓度为29.5±2.9μg/L,较rhbFGF组明显下降(P<0.01)。rhbFGF作用后HLE-B3内Ⅲ型胶原蛋白浓度为1.27±0.29μg/L,较正常组(0.83±0.12μg/L)明显升高,Ele作用后HLE-B3内Ⅲ型胶原蛋白的浓度为0.69±0.11μg/L,较rhbFGF组明显下降(P<0.01)。结论:Ele抑制rhbFGF诱导的HLE-B3增殖的同时也能抑制HLE-B3内Ⅰ,Ⅲ型胶原蛋白合成,干扰HLE-B3纤维化,可望成为防治后囊膜混浊的理想药物。AIM: To investigate the effects of elemene (Ele) on collagen synthesis of human lens epithelial cells 133 (HLE-B3). METHODS: Using 10μg/L recombinant human basic fibroblast growth factor( rhbFGF) to induce proliferation of HLE-B3. Proliferative HLE-B3 were incubated with 80mg/L Ele in CO2 incubator for 24 hours . Then The contents of collagen Ⅰ and collagen Ⅲ of HLE-B3 were assayed by enzyme-linked immunosorbent assay(ELISA). RESULTS: The collagen Ⅰcontent of HLE-B3 was 53.5 ± 5,4μg/L in rhbFGF group, which increased obviously compared with control group (38. 5 ± 2. 3μg/L), The collagen Ⅰ contents of HLE-B3 was 29.5 ±2.9μg/L in Ele, which decreased obviously to compare with rhbFGF group ( P 〈 0, 01 ) ; The collagen Ⅲ content of HLE-B3 was 1,27 ± 0, 29μg/L in rhbFGF group,which increased obviously com- pared with control group (0.8 ± 0. 1μg/L). The collagen contents of HLE-B3 were 0. 7 ± 0. 1μg/L in Ele groups, which decreased obviously to compare with rhbFGF group(P〈0.01 ). CONCLUSION: Ele could effectively inhibit HLE-B3 proliferation induced by rhbFGF. Ele could inhibite the synthesis of collagen Ⅰ and collagen Ⅲ in HLE-B3 as well. The inhibitive effects of Ele on the HLE-B3 proliferation and the synthesis of collagen Ⅰ and collagen make it probably become reliable and effective drugs to prevent and treat posterior capsular opacification.
