摘 要:通过比较拟南芥和蓖麻的SAD基因同源区域,设计引物并以木薯基因组DNA为模板进行PCR扩增,得到长度为240 bp的基因片段,命名为木薯SAD。序列相似性检索结果表明,木薯SAD基因的核苷酸序列与基因库中登陆的大戟科植物蓖麻、千年桐的mRNA序列同源性达94%,与大豆、芝麻、葵花等油料作物的mRNA序列同源性分别达到87%、86%和82%;系统进化分析进一步表明,木薯SAD基因与大戟科植物蓖麻、麻疯树和千年桐的亲缘关系最近,该结果与传统分类学将木薯与蓖麻、千年桐和麻疯树归于大戟科的结论相符,而木薯SAD基因与芝麻、大豆、莲花、葵花等植物种仁不饱和脂肪酸含量较高的植物在进化上具有较高的保守性。By comparing the Arabidopsis and Ricinus SAD gene homology region, primers were designed to cassava genomic DNA as a template for PCR amplification, the size of the 240bp gene fragment were received, named mushu SAD. Through sequence similarity search results showed that its nucleotide sequence with Genbank landing Medium Euphorbiaeeae ricin, Aleurites montana mRNA sequence homology of 94%. With soybean, sesame, sunflower and other oil-producing crops mRNA sequence homologily,respectivelyof 87%, 86% and 82%. Phylogenetic analysis further showed that the cassava gene SAD has the nearest relationship with Euphorbiaceae plant castor, jatropha and Aleurites montana , the result consistent to the traditional taxonomy to Euphorbiaceae in line with Euphorbiaceae plant castor, jatropha and Aleurites montana sesame, has high conservation to soybean, lotus, sunflower kernel ere.high content of unsaturated fatty acids in the evolution of the plant.
