烟酸缓释片体内外释放特征研究

摘　　要：目的研究烟酸缓释片体外释放特征和体内药代动力学特征。方法采用体外释放度测定法和在体犬口服药代动力学参数测定法分别测定药物的体内外释放特征,对犬口服药代动力学特征和体内外释放度特征进行分析。结果烟酸缓释片与普通片比较具有缓释特征,药代动力学分析显示烟酸缓释片体内分布为单室模型,Wagner-Nelson法分析显示药物在体内的吸收呈现先慢后快的特征。结论烟酸缓释片具有缓释和释放先慢后快的特征。Objective To study the release characteristics in vitro and the pharmacokinetic characteristic in vivo of Nicotinic Acid Sustained- Release Tablet. Methods The nicotinic acid release rate was tested by in vitro method and the pharmacokinetic characteristic was tested by in vivo method in dogs. The data were analyzed by pharmacokinetic software. Results Nicotinic Acid Sustained- Release Tablet showed sustained release characteristic compared to the normal release tablet. The distribution model of nicotinic acid in dogs was one compartmental model, the nicotinic acid in the sustained- release tablet released slowly at first phase and then rapidly at second phase using Wagner- Nelson analysis. Conclusion Nicotinic Acid Sustained- Release Tablet shows the pharmacokinetic characteristics of sustained release and slow to rapid release.
