摘  要:目的研究γ射线对人外周血淋巴细胞Cx43和Egr-1基因转录表达的影响。方法对数生长期的淋巴细胞,分别给予2、4和6Gy的^(60)Coγ射线照射,并于照射前(对照组)和2Gy照射后4、8、12、24、48和72h及不同剂量照射后12h,分别提取总RNA,反转录成cDNA。利用实时荧光定量PCR技术,检测各组Cx43和Egr-1基因表达改变。结果人外周血淋巴细胞Cx43mRNA表达水平在2Gy照射后4~12h明显增高,为对照组的6.74倍以上(P<0.05);24~72h,其表达水平与对照组相比没有明显变化。不同剂量照射后12h,2Gy和6Gy时表达水平高,是对照组的10.52倍以上(P<0.05)。Egr-1mRNA表达水平在2Gy照射后的8h和24h增高,但没有统计学意义;4、12、48和72h,Egr-1mRNA表达水平比对照组显著降低(P<0.05)。不同剂量照射后12h,2Gy时,Egr-1mRNA表达水平比对照组降低(P<0.05),4Gy时,其表达水平接近对照组,6Gy时,表达水平增高,为正常对照组的7.94倍(P<0.05)。结论γ射线照射后能够明显上调Cx43基因表达。Egr-1要达到表达水平增高倍数和Cx43倍数相同,所需剂量比Cx43大。Objective To study the effect of γ-rays on the transcription of Cx43 and Egr-1 gene in human peripheral blood lymph cells. Methods Lymph cells in logarithmic growing phase were irradiated by γ-rays of 2, 4, 6 Gy respectively. RNA were extracted before irradiation, 12 hours after irradiation and also 4, 8, 12, 24, 48, 72 hours after 2 Gy irradiation. The RNA turned into cDNA by inverse transcription. The expression of Cx43 and Egr-1 mRNA were measured using fluorescent quantitative PCR. Results The level of Cx43 mRNA in lymph cells increased significantly at 4-12 h after 2 Gy irradiation and more than 6. 74 fold compared with the control(P〈0. 05). At 24-72 h, the expression level was not significantly different from the control. 12 hours after irradiation the level of Cx43 expression increased significantly in 2 and 6 Gy irradiation and was above 10. 52 fold compared with the control. The level of Egr-1 mRNA expression increased but not statistically significantly at 8 and 24 h in 2 Gy irradiation and decreased at 4, 12, 48, 72 h compared with the control(P〈0. 05). 12 hours after irradiation , the level of Egr-1 mRNA expression significantly decreased in 2 Gy irradiation compared with the control, (P〈0. 05), close to the control in 4 Gy irradiation, and increased significantly and was 7. 94 fold compared with the control (P〈0. 05). Conclusion γ-rays can significantly upregulate the expression of Cx43 mRNA. In order to get the same expression level of Cx43, the irradiation dose that Egr-1 needs is higher than that of Cx43.
