低氧环境中几种中药注射液对大鼠骨髓间充质干细胞增殖的影响

摘　　要：目的:探讨在低氧环境下雪莲、红花和红景天注射液对大鼠骨髓间充质干细胞(rMSCs)增殖的影响。分离、纯化、培养大鼠rMSCs细胞,选择雪莲、红花和红景天注射液,通过四甲基偶氮唑法检测法(MTT)和Brdu标记的方法检测这些中药注射液在低氧条件对大鼠rMSCs增殖的促进作用。结果:在低氧条件下红景天注射液对rMSCs的增殖效率与对照组比较差别有显著性,有明显的增殖作用。而红花注射液对rMSCs的增殖无显著作用。雪莲注射液对rMSCs的增殖率有明显的抑制作用。结论:红景天注射液在低氧条件下具有促进rMSCs增殖的作用。雪莲注射液在低氧条件下具有抑制rMSCs增殖的作用。Objective.To study whether several natural traditional chinese medicines could affect the proliferation of bone marrow mesenchymal stem cell of rat （rMSCs） under hypoxic condition. Methods. We isolated and cultured the rMSCs under hypoxic condition, and Carthamus tinctoriusL , Saussurea involuerata and Rhodiola kirilowii injections were selected in this study. The proliferate effects of these injections on rMSCs was detected by MTT and BrdU labeling approach. Results：The proliferation of the Rhodiola kirilowii injections on rMSCs was significantly enhanced as compared with the control group （P〈0. 05） but not change for Carthamus tinetoriusL injection, in contrast, the proliferation of rMSCs was significantly reduced by the Saussurea involucrata injection. Conclusions. The Rhodiola kirilowii injection can promote the proliferation of rMSCs. The Saussurea involucrate injection can inhibit the proliferation of rMSCs.
