摘  要:对兔胚胎干细胞核移植过程中受体卵子的去核方法、供核兔胚胎干细胞的预处理及电融合的场强等操作技术进行了比较试验。结果表明,压迫式去核方法的去核有效率达85.71%,略高于注吸式。当电融合场强为1.6或1.8 kV/cm时,重组胚的卵裂率和桑、囊胚率较高,分别达到33.3%和16.67%。在操作液中添加PVA对獭兔干细胞核转入融合与后期囊胚发育均有不同程度的影响,使囊胚发育率显著提高。将11个融合胚及5枚发育囊胚移植到受体兔体内,未发现妊娠现象。Different enulaeation methods, pretreatment of rabbit donor embryonic stem cells and the different field intensities of electric fusion in the process of nuclear transfer were studied and compared. The results showed that the enucleation rate of oocytes by the method of extrusion was 85.71%, a httle better than the method of absorb-infusion. When the electric field intensity was 1.6 or 1.8 kV/cm, the cleavage rate and the morula/blastocyst rate were higher than other treatments, reaching 33.B% and 16.67%, respectively. Moreover, the rate of morula and blastocyst increased when PVA was added to the operation medium. No pregnancy was observed when 11 reconstructed embryos and 5 reconstructed blastocysts were transferred into the receptor rabbit.
