摘  要:以鲫鱼Carassius auratus Linn.为试验鱼类,研究了其暴露于不同质量浓度的2,2’,4,4’-四溴联苯醚(PBDE-47)和十溴联苯醚(PBDE-209)后鱼肝微粒体中CYP3A1和GST酶活性的动态变化。结果表明,鲫鱼在0.10~5.00mg·L-1的PBDE.47和5.00~50.0mg·L0的PBDE-209中暴露15d后,除0.10mg·L-1 PBDE.47、5.00mg·L-1和10.0mg·L-1PBDE-209试验组外,其余各试验组的鱼肝微粒体中CYP3A1被诱导,呈显著的剂量-效应关系。CYP3A1的活性随着作用时间的延续而上升,到试验第15天时达到最高,但上升速率最快的阶段为试验的第0-5天。而GST酶则表现出不同的特点,呈先升高后降低的趋势。经过15d试验,除0.10mg·L-1PBDE-47和5.00mg·L-1PBDE-209以外的各试验组鲫鱼肝脏微粒体中的GST活性仅为对照组的12.74%~85.35%,表明PBDEs已对鱼体肝脏GST产生了较为严重的影响。研究表明,鱼类肝脏微粒体中CYP3AI和GST酶可作为污染生物标志物来评价PBDEs的早期污染毒理效应。The present paper intends to introduce the authors' experimental research on the effects of tetrabromodiphenyl ether (PBDE-47) and decabromodiphenyl ether(PBDE-209) on the activities of CYP3A1 and GST in the liver microsome of Carassius auratus Linn.exposed to PBDE-47 and PBDE-209. For the research purpose, we had put the fishes to water with the concentration of PBDE-47 being 0.10-5.00 mg·L-1 and PBDE-209 being 5.00-50.0 mg·L-1, then isolated the liver microsome of the fishes to be tested. The CYP3A1 and GST were analyzed after 5-15 day exposure at the temperature of 22℃. The results of our experiments proved that the normal values of C YP3 A 1 and G ST was separately (1.034-0.03) U-rag 1 and (1.57±0.02 ) U·mg-1. And the test groups of 0.10mg·L-1 PBDE-47, 5.00 mg·L-1 and 10.0 mg·L-1 PBDE-209 had no effects on CYP3A1 in the liver microsome after treated 15 days(P〉0.05). But the other test groups could cause the obvious changes to CYP3A1 (P〈0.01). During 15 days, with the concentra- tion of PBDEs increased, CYP3A1 tended to increase, which turned to prove CYP3A1 and PBDEs concentrations were closely re- lated. The activities of CYP3A1 in the test groups could reached the highest value at the 15th day, and they raised fastest within the first 5 days. The results also showed that 0. l 0mg·L-1 PBDE-47 and 5.00mg·L-1 PBDE-209 had no effects on GST compared with the control group (p〉0.05). But the other test groups could cause the obvious change to GST. At the first 10 days, GST increased, then it began to decrease .The activities of GST in these tested groups were only 12.74-85.35% of them in the control group. The results indicated that CYP3A1 and GST in the liver microsome of fishes could be used as the better biomarkers to estimate the pollution effect and toxicity of the PBDEs.
