摘 要:目的观察鹿茸多肽对大鼠关节软骨细胞体外传代培养中出现的去分化现象的作用。方法将第3代软骨细胞分为空白对照组、鹿茸多肽不同浓度组、硫酸氨基葡萄糖不同浓度组传代,使之进入第4代,同时以第2代软骨细胞为对照组,进行阿力新蓝染色法检测GAG(glycosaminoglycan)含量和结构,RT-PCR(reverese transcript-polymerase chain reaction)法检测Ⅱ型胶原、Aggrecan蛋白,对鹿茸多肽抗软骨细胞去分化进行分子生物学研究。结果鹿茸多肽对软骨细胞胞外基质中GAG、Ⅱ型胶原和Aggrecan蛋白表达有显著促进作用(P<0.01),其作用优于硫酸氨基葡萄糖。结论鹿茸多肽对体外培养的软骨细胞去分化现象有抑制作用,其作用优于硫酸氨基葡萄糖。Objective To observe the function of pilose antler polypeptides (PAP) on dedifferentiation of rat ehondrocyte subeuhivated in vitro by means of PAP interfering and controled experiment. Methods The 3rd passage chondrocytes were divided into blank group, different concentration PAP groups, which were passaged to 4th generation and contrast to the 2nd passage group.The chondrocytes of different groups were detected with the method of alcian blue test for the content and constructure of glycosaminoglycan (GAG), reverese transcript- polymerase chain reaction (RT-PCR) for type Ⅱ collagen and Aggreean, to observe PAP's function regarding to the appearance and functional status in the process of chondrocyte's senescence. Results PAP significantly enhance the content of GAG, type Ⅱ collagen and Aggrecan of ECM (P〈0.01). PAP has anti-dedifferentiation function that was obviosly better than GS.Conclusion PAP can significantly inhibit rat chondrocyte dedifferentiation occurred in subcultivation.
