摘  要:目的探讨肝星状细胞钾通道对肝星状细胞增殖的影响,观察钾通道阻滞剂四乙胺(tetraethy-lammonium,TEA)对肝星状细胞增殖和钾离子电流的作用。方法以TEA作用于传代培养的肝星状细胞,通过MTT法、流式细胞术及单通道细胞膜片钳技术,分别检测TEA对肝星状细胞的增殖、细胞周期及细胞膜钾通道的影响。结果TEA对肝星状细胞的增殖抑制效应具有时间依赖与剂量依赖的特点,使细胞周期阻滞于G0/G1期,单通道细胞膜片钳发现肝星状细胞上有大电导离子通道存在,其电导值为(210.86±5.49)pS(n=7),TEA可明显减少外向该离子通道开放概率,TEA0.0、2.5、5.0、10.0、20.0和40.0mmol/L组的开放概率为(0.327±4.80)、(0.316±4.38)、(0.279±1.72)、(0.245±1.21)、(0.194±1.13)和(0.118±1.62)(F=22.561,其中10.0、20.0和40.0mmol/L组分别与0.0mmol/L组比较,差异有统计学意义(P<0.01)。结论肝星状细胞上有大电导钾通道存在,阻断钾通道可抑制肝星状细胞增殖,钾通道对细胞增殖可能具有调控作用。[Objectives] To investigate the role of potassium channels in proliferation of hepatic stellate cells (HSCs) through observing the effects of tetraethylammonium (TEA) on proliferation and outward potassium currents in hepatic stellate cells (HSCs). [Methods] Hepatic stellate cells (HSCs) were treated with TEA. The effect of TEA on proliferation of hepatic stellate cells (HSCs) was assessed by MTT assay. Cell cycle were detected by flow cytometry. The potassium currents were recorded by patch clamp technique. [Results] TEA inhibited the proliferation of HSCs in dose and time dependent manners. The cell cycle was arrested at GO/G1 phase after treatment with TEA. Potassium channels were detected by patch-clamp in the activated HSCs studied and showed a slop conductance of (210.86 ± 5.49) pS (n =7). Exposure of HSCs to 0 mmol/L -40 mmol/L TEA reduced the open probability (NPo) out- ward potassium currents significantly from (0.327 ± 4.80) to (0.118 ± 1.62) (P 〈0.01). [Conclusions] Activated HSCs in rat contain large-conductance potassium channels. Blocking potassium channels could inhibit proliferation of HSCs. Channels play an important role in regulating proliferation of HSCs .
