摘　　要：目的探讨抵抗素样分子α（RELMa）在ApoE基因敲除鼠动脉粥样硬化斑块内的表达及其对血管平滑肌细胞（VSMC）增殖、迁移的影响。方法C57BL／6JApoE基因敲除鼠及C57BL／6J野生型小鼠各9只，喂食高脂饲料，24周后处死，自主动脉根部至腹主动脉离断血管，石蜡包埋血管后作连续切片，行HE染色及RELMa免疫组化，RT-PCR检测斑块内RELMα mRNA表达，用不同浓度的重组RELMα刺激培养的VSMC，观察VSMC增殖及其迁移情况，采用SPSS11．0统计软件包，以one-way ANOVA分析各组之间的差异。结果ApoE基因敲除鼠高脂饲养24周后，主动脉根部形成动脉粥样硬化斑块，免疫组化及其RT-PCR可见RELMα蛋白及其mRNA在粥样硬化斑块内明显表达，野生型C57BL／6J鼠血管壁内，未见RELMα及其mRNA表达，重组RELMα能刺激体外培养的VSMC增殖（RELMα刺激组cpm分别为[（2811．21±216．89），（4056．87±220．65），（5061．45±335．86），与对照组（1609．58±203．53）比较，P〈0．01]重组RELMα也能刺激体外培养的VSMC迁移[RELMα刺激组迁移细胞数分别为（130．54±12．98），（158．39±11．58），（203．50±17．37），与对照组（70．54±11．92）比较，P〈0．01]。结论RELMα在动脉粥样硬化斑块内表达，其能促进VSMC增殖及迁移。Objective To explore the expression of resistin-like molecules-α （RELMct） in athernsclerotic plaque of ApoE-/- mouse, and to study the effects of RELMa on the proliferation and migration of vascular smooth musclee cells （VSMCs）. Method Nine ApoE-/- mice and nine C57BL/6J mice were fed with high fat diet. All mice were sacrificed 24 weeks after force feeding. Vessels were dissected from to abdominal aorta. Sections of aortic tissue were stained with HE dyeing and RELMa in aortic tissue was assayed by immunohistochemistry. The expression of RELMα mRNA in vessels was detected by RT-PCR. The effects of different concentration RELMα in different concentrations on the proliferation and migration of VSMCs were detected. Data was expressed as mean± standard deviation. ANOVA were used for comparison in SPSS 11.0, and changes were considered as statistically significant if P value was less than 0.05. Results Atherosclerosis plaque formed in aortic root of ApoE-/- mice after they were fed with high fat diet for 24 weeks. RELMa protein and RELMα mRNA were found by immunohistochemistry and RT-PCR in atherosclerotic plaque of ApoE-/- mouse. RELMα protein didn＇t be found in vessels of control mouse. RELMα promoted the proliferation of VSMCs （RELMa groups： 2811.21 ± 216.89,4056.87± 220.65,5061.45±335.86,vs. control 1609.58±203.53, P 〈 0.01）. RELMα promoted the migration of VSMCs （RELMa groups： 130.54± 12.98,158.39± 11.58,203.50± 17.37 vs. control：70.54± 11.92, P 〈0.01）. Conclusions RELMα expresses in atherosclerotic plaque of ApoE-/- mouse. RELMα enhances the proliferation and migration of VSMCs of aorta.
