酸味中药复方对2型糖尿病大鼠主动脉AGEs含量及受体基因表达的影响

摘　　要：目的观察酸味中药复方对2型糖尿病(T2DM)大鼠主动脉晚期糖基化终产物(AGEs)含量及受体(RAGE)基因表达的影响,探讨其防治糖尿病大血管病变的机制。方法采取腹腔注射链脲佐菌素(STZ)配合高热量饮食的方法建立T2DM动物模型。将实验用SD大鼠随机分为正常组、模型组、氨基胍(AG)组及中药组。在给药8周和12周分别采用葡萄糖氧化酶法测大鼠空腹血糖(FBG),荧光法测大鼠主动脉AGEs及胶原含量,Real-timePCR法检测大鼠主动脉RAGE基因表达。结果T2DM大鼠FBG、主动脉AGEs含量及胶原含量、主动脉RAGE基因表达水平均明显高于正常组(P<0.05);中药组大鼠FBG、主动脉AGEs及胶原含量、主动脉RAGE基因表达水平均明显低于模型组(P<0.05)。结论酸味中药复方可能通过抑制糖尿病大鼠主动脉AGEs形成和下调RAGE基因表达实现其防治糖尿病大血管病变目的。Objective To investigate the possible mechanism of compound Chinese sour taste herbs （CS） in preventing and ameliorating diabetic macroangiopathy by analyzing the effects of CS on the deposition of advanced glycation end products （AGEs） and gene expression of receptor for advanced glycation end products （RAGE） in the aorta tissue of rats with type 2 diabetes mellitus （T2DM）. Methods Rat model of T2DM was established by peritoneal injection of streptozotocin （STZ） and high caloric diet feeding. Experimental SD rats were divided into the normal group, the model group, the aminoguanidine （AG） group, and the CS group. At the end of the 8th and 12th week, the fasting blood glucose （FBG） was measured by glucose oxidase method; content of AGEs and collagen in aorta detected by fluorescent method and gene expression of RAGE in aorta determined by Real-time PCR method. Results FBG, AGEs＇and collagen contents and RAGE expression in aorta of model rats were all higher than those in the normal control group （ P 〈 0.05）, while all these indices were lower in the CS group than in the model group （P 〈0.05）. Conclusion CS could realize the goal for preventing and ameliorating diabetic macroangiopathy by way of suppressing the production of AGEs and down-regulating the gene expression of RAGE in aorta of T2DM rats.
