五味子总RNA提取方法的建立

摘　　要：五味子是一种多糖含量很高的木本植物,由于多糖的干扰,其RNA制备比较困难。研究以五味子的叶片为材料,建立了一种稳定的从五味子中分离总RNA的方法。该方法以CTAB为基本提取试剂,通过酚、仿抽提纯化,最后应用LiCl来沉淀RNA,可以在1h内提取出纯度高的RNA。RNA质量可以满足cDNA文库构建、RT-PCR等多种分子生物学研究要求。Schisandra chinesis is a woody plant which had high level of polysaccharide. Due to the interference of polysaccharide, it is difficult to isolate RNA from Schisandra chinesis. In the present study, a method for RNA isolation from leaves of Schisandra chinesis was build. This method ia a kind of CTAB method, using phenol and chloroform to purify RNA, and RNA was deposited by LiCl, The entire experiment can be completed in 1h. The quantities of RNA can meet the request of many experiments, such as cDNA library construction and RT-PCR reaction.
