蛴螬提取物诱导人肺癌A549细胞凋亡的机制研究

摘　　要：目的:观察蛴螬提取物对人肺癌A549细胞增殖的影响及其机制。方法:采用MTT法检测蛴螬提取物对人肺癌A549细胞的生长抑制率;用HE染色方法观测凋亡细胞的形态学变化;运用免疫组织化学SP法检测用药前后细胞周期蛋白D1(CyclinD1)的表达改变。结果:用蛴螬提取物处理的人肺癌A549细胞生长抑制率明显高于对照组;施药早期细胞出现凋亡形态学变化;A549细胞经蛴螬提取物处理后CyclinD1表达均下降。结论:蛴螬提取物在体外对人肺癌A549细胞株有显著的增殖抑制作用,其机制与下调CyclinD1的表达有关。Objective：To observe the proliferation inhibitive effect of Grub Extract on human lung cancer A549 cell line. Methods ： MTT assay was used to measure the growth inhibition rate of human lung cancer A549 cell line treated with the Grub Extract. further testing the changed morphology by HE and the expression of CyclinD1 was detected by immunohistochemistry before and after exposure to the Grub Extract. Results ：The growth inhibition rate of A549 cells treated with the Grab Extract was significantly higher than that of control. The showed the apoptesis changes by Grub extract before 24hours and The expression of CyclinD1 was down-regulated. Conclusion：The proliferation of human lung cancer A549 cell line can be inhibited significantly by the Grub Extract in vitro. It is related with down-regulation of CyclinD1.
