黄芪莪术配伍药物血清对胃癌MKN45细胞增殖和分化的影响

摘　　要：目的:观察黄芪、莪术配伍药物血清对人胃癌细胞株MKN45增殖和分化的影响,比较中药配伍和单味药应用的不同,为益气活血方法治疗胃癌提供实验依据。方法:通过中药血清药理学方法制备黄芪、莪术配伍药物血清,采用MTT法检测药物血清对MKN45细胞的增殖抑制率;酶标仪测定分化标志酶AKP、LDH的活性。结果:MTT法实验中,莪术组作用48h后对细胞的抑制作用与正常组相比有显著意义(P<0.01),作用72h,莪术组、配伍组对细胞增殖仍有抑制作用,与正常组相比有显著意义(P<0.01),且配伍组在24h、48h、72h均对MKN45细胞增殖维持稳定的抑制率。通过酶标仪检测,配伍组作用的MKN45细胞AKP、LDH活性表达均明显降低,与正常组相比有显著意义(P<0.05),配伍组与黄芪组相比有统计学意义(P<0.01)。结论:黄芪、莪术配伍药物血清能够抑制胃癌MKN45细胞增殖并诱导其分化,与单味药应用相比具有增效、疗效稳定优势,提示益气活血法是中医药治疗胃癌的有效治则治法。Objective ： To observe the effect of drug serum of compatibility Astragalus and Curcumae on proliferation and differentiation of human gastric cancerous cell strain MKN45, compare differences in application of drug combination and individual, and provide experimental evidence for treating gastric cancer by invigorating qi and activating blood. Methods ： Preparing drug serum of compatibility Astragalus and Curcumae according to pharmacology of Chinese materia medica, detecting proliferation inhibition rate of MKN45 cells by MTr,and determining the activity of marker enzymes AKP,LDH by an enzyme - labeling instrument. Results：In the MTT experiment, Group Curcumae inhabited ceils more powerfully after 48 hous in contrast with Control Group（P 〈 0. 01 ）. After 72 hours, Group Curcumae and Combination Group still inhibited cell proliferation, displaying the difference between them and Control Group had statistical significance（P 〈 0. 01 ）. Combination Group stably inhibited MKN45 proliferation after 24 hours ,48 hours and 72 hours. By an enzyme-labeling instrument, activity expression of AKP, LDH of MKN45 in Group Curcumae and Combination Group decreased markedly, displaying the difference between them and Control Group had statistical significance（ P 〈 0. 05） and the difference between Combination Group and Group Astragalus had statistical significance（ P 〈 0. 01 ）. Conclusion ： Drug serum of compatibility Astragalus and Curcumae inhibited proliferation and induced differentiation of gastric cancerous ceil strain MKN45. Comparing with individual drugs, the combination had the advantage of effect stability and synergy, which proves that the principle of invigorating qi and activating blood is effective for treating gastric cancer by TCM.
