醒脑开窍针刺法主穴内关对脑缺血模型(MCAO)大鼠脑神经细胞坏死的抑制作用研究

摘　　要：目的:分别从光镜和电镜两个角度探讨内关穴与非穴对脑缺血模型(MCAO)大鼠神经细胞坏死的抑制作用。方法:参照Zea-Longa线拴法复制大鼠大脑中动脉缺血模型(MCAO),分别对"醒脑开窍"针刺法主穴内关以及非穴(胁下非经非穴点),施以频率180次/s,持续时间5s的针刺干预,以光镜下神经细胞坏死率及电镜下神经细胞坏死程度作为评价效应指标。结果:光镜下,与模型对照组相比,非针刺组和非穴组神经细胞坏死率均无统计学意义(P>0.05),而内关组的神经细胞坏死率明显降低,且有统计学意义(P<0.05)。电镜下,非针刺组和非穴组神经细胞超微结构接近模型对照组,表现为神经细胞的高度水肿及细胞器的严重破坏;内关组则接近正常组和假手术组,神经细胞破坏程度均较低。结论:针刺内关可抑制MCAO大鼠神经细胞坏死,保护神经元,具有穴位特异性。Objective ： To observe the effect of Nei-guan PC6 and nonacupoint on inhibiting cerebral nerve cell necrosis beared by rat model of middle cerebral artery occlusion （MCAO） with light microscope and electron microscope. Method ： The rat model of middle cerebral artery occlusion （MCAO） was established by using thread according to Zea-Longa. Then acupuncture the main acupoint （Neiguan PC6） and nonaeupoint （below the costal region） guided by theory of waking up a patient from unconsciousness （XNKQ） for 5 seconds by 180 times / second. Finally, measure the rate of cerebral nerve cell necrosis under the light microscope and the degree of cerebral nerve cell necrosis under the electron microscope to estimate the effect. Result： Compared to control group, there was no significant difference between non-acupuncture group and nonacupoint group on cerebral nerve cell necrosis rate under light microscope （ P 〉 0.05 ）, while the rate was decreased in Nei-guan PC6 group （ P 〈 0.05 ）. Under the electron microscope, ultramierostructure in nonacupuncture group and nonacupoint group got close to the one in control group, the nerve cell edema was serious and cell organs were destroyed seriously. The nerve cells were less destroyed in Nei-guan PC6 group, and the degree was near to the normal group and sham operation group. Conclusion： Acupuncture on Nei-guan PC6 can inhibit MCAO rats＇ cerebral nerve cell necrosis, and protect nerve cells. It shows acupoint specificity.
