摘　　要：目的建立HPLC法测定目安眼膏在家兔房水内阿昔洛韦(ACV)含量的方法,为目安眼膏在兔眼内转运动力学研究提供定量方法。方法房水样品用25%高氯酸沉淀蛋白后用Na2CO3调pH值至近中性,离心取上清液测定。色谱条件:Phenomenex C18柱(250 mm×4.6 mm,5μm);流动相为甲醇-水(10∶90);流速为1.0 mL/min;柱温为30℃;检测波长为254 nm。结果房水样品中阿昔洛韦浓度在0.037～2.78μg/mL的范围内呈良好的线性关系,低、中、高3种浓度的平均提取率为102.3%(RSD=3.5%)、98.4%(RSD=1.9%)和101.5%(RSD=2.2%)。结论该方法专属性强、灵敏度高、结果准确,可用于测定目安眼膏在兔房水内的阿昔洛韦浓度。Objective To set up a method to determine the concentration of Acyclovir （ACV） in Muan eye ointment with HPLC for the study of the ointment pharmacokinetics in rabbit aqueous humor. Methods The protein of sample aqueous humor was precipitated by 25% perchloric acid, its pH value adjusted to neutral, and the aliquot of the supernatant analyzed with HPLC. The chromatography conditions were Phenomenex C18 （250 mm ×4.6 mm, 5 μm）, methyl alcohol-water （10：90）, flow rate 1.0 mL/min, column temperature 30 ℃ and UV detector 254 nm. Results Good linearity was in the range from 0.037 μg/mL to 2.78 μg/mL of Acyclovir concentration in aqueous humor. The average extraction rate were 102.8%, 99.7% and 100.3% in high, middle and low concentration of ACV. Conclusion This method is selective in specificity, sensitive and accurate for the determination of Acyclovir in Muan eye ointment in aqueous humor of rabbits.
