虎杖提取物对小鼠肠运动功能和家兔离体肠平滑肌的影响

摘　　要：目的;观察虎杖提取物对正常小鼠和便秘小鼠大、小肠运动功能及对家兔离体肠平滑肌的影响,并探讨其作用机理。方法:采用在体方法观察大、中、小3个剂量的虎杖提取物对正常小鼠和便秘小鼠大、小肠运动功能的影响;采用离体方法观察大、中、小3个剂量的虎杖提取物对家兔离体肠平滑肌的影响及其作用机理。结果:大、中、小3个剂量的虎杖溶液均能提高正常小鼠和便秘小鼠的小肠墨汁推进百分率,可缩短便秘小鼠首次出现含墨汁粪便的潜伏期,增加6h内排出的含墨汁粪便的颗粒总数;虎杖提取物对家兔离体肠平滑肌的作用与虎杖提取物的浓度和剂量有关,中剂量虎杖水溶液0.2mL对家兔离体肠平滑肌有较明显的兴奋作用,可使肠平滑肌收缩,并能对抗阿托品所致的肠肌舒张,加强乙酰胆碱的收缩肠肌的作用、但对氯化钡,酚妥拉明所致的肠肌收缩影响不大,对肾上腺素所致的肠肌舒张也影响不大,对组胺收缩肠肌有降低的作用。结论:大、中、小3个剂量的虎杖溶液对正常小鼠和便秘小鼠大、小肠蠕动均有促进作用;并可收缩兔离体肠平滑肌的作用,可能和兴奋M受体有关。Objective： Observation of Polygonum euspidatum extract on normal mice and mice constipation, intestinal motor function,explore its removal;Observation of Polygonurn cuspidatum extract rabbit intestinal smooth muscle in vitro, and to investigate the effect of its removal mechanism. Methods ： In vivo method of large, medium and small three doses of Polygonum cuspidatum solution of normal mice and mice constipation, intestinal motor function ; In vitro methods used to observe large ,3 small dose of Polygonum cuspidatum solution of rabbit intestinal smooth muscle in vitro effects. Results： large,3 small dose of Polygonum cuspidatum solution could improve constipation normal mice and mice intestine ink promote percentage constipation can be shortened first mouse droppings containing ink latency, increase within six hours to collect stool samples containing ink particles total; Polygonurn cuspidatum extract on the rabbit intestinal smooth muscle in vitro and the role of Polygonum cuspidatum extract concentration and dose-related,dose Huzhang solution（0.2mL） can significantly promote rabbits in vitro intestinal smooth muscle contraction, and confrontation by atropine, Epinephrine-induced intestinal muscle relaxation to acetylcholine and histamine, barium chloride, phentolamine caused by intestinal muscle contraction performance facilitating role. Conclusion：Large, medium and small three doses of Polygonum cuspidatum solution of normal mice and mice constipation, peristalsis of the small intestine were promoted;Polygonum cuspidatum extract promotes peristalsis, may be Polygonum cuspidatum extract direct access this role role in intestinal smooth muscle cells caused.
