摘 要:目的:建立黄芪有效部位多糖的含量测定方法。方法:用优化后的3,5-二硝基水杨酸(DNS)比色法测定黄芪有效部位多糖的含量。结果:DNS试剂的用量为2ml,显色温度为100℃,显色时间为5min,在500nm处多糖含量与吸光度有良好的线性关系,平均回收率为100.6%,RSD=1.79%(n=5)。结论:该方法操作简便,测定结果准确,重复性好,可用于测定黄芪多糖的含量。Objective To establish a method for the determination of polysaccharides in Astragalus membranaceus.Method Using optimized DNS colorimetric method to determine the polysaccharides.Result The conditions were as follows:3,5-dinitrosalicylic acid 2.0ml,aqueous bath temperature 100℃,and active time 5min.There was a good liner relationship between the concentration of polysaccharides and the absorbance at the wavelength of 500nm.The average recovery was 100.6%,RSD=1.79%(n=5).Conclusion The method is convenient, accurate with good reproducibility and can be used for the determination of polysaecharides in Astragalus membranacells.
